Abstract
Jeruk keprok lokal Indonesia merupakan salah satu jenis jeruk yang sangat digemari karena ukurannya buah besar, aroma buah khas, kulit buah tebal, warna kulit buah yang menarik, serta kulit buah yang mudah dilepas. Namun, jeruk keprok memiliki banyak sekali penyakit dan hama yang dapat mengurangi produksi tanaman jeruk keprok. Untuk itulah, pengendalian hayati diperlukan, salah satunya adalah dengan pemanfaatan bakteri endofit. Salah satu kemampuan bakteri endofit dalam menjadi pengendali hama tanaman pada jeruk keprok adalah dengan memproduksi enzim protease, yang memiliki kemampuan untuk memecah ikatan peptida pada protein pada dinding sel dan cangkang hama. Oleh sebab itu, kerusakan dinding sel akan menyebabkan pertumbuhan hama dan patogen terhambat. Sehingga, enzim protease memiliki potensi sebagai agen biokontrol pada patogen dan hama tanaman. Karena itulah, penelitian ini dilakukan untuk mengisolasi dan mengkarakterisasi bakteri endofit penghasil enzim protease yang diharapkan dapat memiliki potensi sebagai sumber enzim protease. Tahapan dari penelitian ini berupa: (1) Isolasi bakteri dari daun, batang, akar, dan kulit buah; (2) seleksi bakteri endofit proteolitik; (3) Karakterisasi isolat bakteri endofit proteolitik; (4) dan uji aktivitas enzim protease dari ekstrak kasar yang dihasilkan oleh bakteri endofit penghasil enzim protease. Berdasarkan hasil penelitian, ditemukan enam kultur, yaitu isolat LB21b dan LB22a dari batang, LA23a dari akar, LK11b dan LK11c dari kulit buah, dan LD11a dari daun. Berdasarkan hasil karakterisasi yang dilakukan, diduga bahwa isolat LB22a adalah Pseudomonas sp., isolat LA23a adalah Enterobacter sp., LK11b adalah Klebsiella sp., dan LD11a adalah Escherichia sp. Namun, isolat LK11c dan LB21b tidak berhasil dikarakterisasi. Selain itu, dapat diketahui bahwa isolat LB21b memiliki nilai aktivitas enzim protease tertinggi, yaitu sebesar 2,2370 U/mL pada jam inkubasi ke-48.
English Abstract
Indonesian local tangerines are one of the most popular types of oranges because of their large fruit distinctive fruit aroma, thicker fruit skin, attractive fruit skin color, and their fruit skin is easy to peel. However, tangerines have many diseases and pests that can reduce the production. For this reason, biological control is needed, and one of which is the use of endophytic bacteria. One of the abilities of endophytic bacteria in controlling tangerines pests is by producing protease enzymes, which have the ability to break down peptide bonds in proteins in cell walls and shells of pests. Therefore, damages made by the protease enzymes to the cell wall will inhibit the growth of pests and pathogens. Thus, protease enzymes have potential as biocontrol agents in plant pathogens and pests. Therefore, this research was conducted to isolate and characterize endophytic bacteria that produce protease enzymes which are expected to have potential as a source of protease enzymes. The stages of this research were: (1) Isolation of bacteria from leaves, stems, roots, and fruit skins; (2) selection of proteolytic endophytic bacteria; (3) Characterization of proteolytic endophytic bacteria isolates; (4) and test the activity of protease enzymes from crude extracts produced by endophytic bacteria producing protease enzymes. Based on the results of the study, six cultures were found, namely isolates LB21b and LB22a from stems, LA23a from roots, LK11b and LK11c from fruit skins, and LD11a from leaves. Based on the results of the chemical identification test, it was suspected that isolate LB22a was Pseudomonas sp., isolate LA23a was Enterobacter sp., LK11b was Klebsiella sp., and LD11a was Escherichia sp. However, isolates LK11c and LB21b were not characterized. In addition, it can be seen that isolate LB21b has the highest protease enzyme activity value, which is 2.2370 U/mL at 48 hours of incubation.
