Abstract

Pisang adalah komoditi pangan ke empat terpenting di dunia setelah beras, susu dan gandum. Pisang di Indonesia merupakan komoditi pertanian dengan produksi paling tinggi di antara buah-buahan lainnya. Namun masih terdapat faktor pembatas yang menjadi kendala dalam peningkatan produksi tanaman pisang yaitu adanya serangan penyakit darah yang disebabkan oleh bakteri patogen Ralstonia syzygii subsp. celebesensis, sehingga perlu dilakukan pengendalian yang tepat dengan memanfaatkan agens hayati seperti potensi antagonis dari bakteri rizosfer pada tanaman Pisang Cavendish dan Pisang Kepok. Penelitian dilakukan di Laboratorium Penyakit Tumbuhan Jurusan Hama dan Penyakit Tumbuhan Fakultas Pertanian Universitas Brawijaya, Malang. Penelitian dimulai pada Bulan Desember 2019 sampai dengan September 2020. Pengujian menggunakan Rancangan Acak Lengkap dengan 7 perlakuan yang diulang sebanyak 3 kali. Variabel pengamatan meliputi perhitungan total koloni bakteri, indeks penghambatan zona hambat bakteri antagonis terhadap R. syzygii subsp. Celebesensis dan karakterisasi bakteri. Data pengamatan dianalisis menggunakan analisis ragam anova dan apabila terdapat perbedaan nyata maka dilanjutkan dengan Uji Duncan taraf 5%. Hasil eksplorasi bakteri rizosfer tanaman Pisang Cavendish dan Kepok diperoleh 46 isolat. Sebanyak 22 isolat diperoleh dari rizosfer Pisang Cavendish dan 24 isolat diperoleh dari rizosfer Pisang Kepok. Terdapat 40 isolat yang tidak bersifat patogenik pada pengujian hipersensitif, sedangkan pada hasil seleksi antagonis diperoleh sebanyak 7 isolat bakteri. Sebanyak 5 isolat (C020, K007, K009, K017 dan K019) digunakan untuk uji antagonis lanjutan. Berdasarkan hasil uji antagonis diketahui bahwa isolat dengan kode C020 menghasilkan zona hambat terbesar jika dibandingkan keempat isolat lainnya. Namun zona hambat dari isolat dengan kode C020 tidak lebih besar jika dibandingkan dengan kontrol positif menggunakan bakterisida berbahan aktif Streptomisin sulfat. Hasil identifikasi dan karakterisasi morfologi koloni menunjukkan isolat patogen dan kelima isolat bakteri antagonis berbentuk bulat, isolat patogen memiliki pusat berwarna merah gelap dengan tepian berwarna putih, sedangkan isolat C020 berwarna kuning, isolat K007 dengan K009 berwarna putih dan isolat K017 dengan K019 berwarna putih bening, dan semua isolat tergolong Gram negatif. Berdasarkan hasil identifkasi dan karakterisasi diketahui bahwa isolat C020 merupakan bakteri dari genus Erwinia sp. atau Pantoea sp., sedangkan isolat K007, K009, K017 dan K019 merupakan bakteri dari genus Pseudomonas sp

English Abstract

Bananas are the fourth most important food commodity in the world after rice, milk and wheat. Bananas in Indonesia are an agricultural commodity with the highest production among other fruits. However, there are still limiting factors that become an obstacle in increasing banana plant production, namely the presence of blood disease caused by the pathogenic bacteria Ralstonia syzygii subsp. celebesensis, so it is necessary to carry out proper control by utilizing biological agents such as the antagonistic potential of rhizosphere bacteria in Cavendish Banana and Kepok Banana. The research was conducted at the Laboratory of Plant Diseases, Department of Pests and Plant Diseases, Faculty of Agriculture, Brawijaya University, Malang. The study was started from December 2019 to September 2020. The test used a completely randomized design with 7 treatments that were repeated 3 times. The observation variables included the calculation of total bacterial colonies, inhibition index of the zone of inhibition of antagonistic bacteria against R. syzygii subsp. celebesensis and bacterial characterization. Observation data were analyzed using ANOVA analysis of variance and if there is a significant difference, then proceed with the Duncan test at 5% level. The results of exploration of the rhizosphere bacteria of Cavendish and Kepok banana obtained 46 isolates. A total of 22 isolates were obtained from the Cavendish Banana rhizosphere and 24 isolates were obtained from the Kepok Banana rhizosphere. There were 40 isolates that were not pathogenic in hypersensitivity testing. Meanwhile, the antagonist selection results obtained 7 bacterial isolates. A total of 5 isolates (C020, K007, K009, K017 and K019) were used for further antagonistic testing. Based on the results of the antagonist test, it was known that the isolate with code C020 produced the largest inhibition zone compared to the other four isolates. However, the zone of inhibition of the isolates with code C020 was not larger than the positive control using a bactericide with the active ingredient Streptomycin sulfate. The results of the identification and characterization of colony morphology showed that the pathogenic isolates and the five antagonistic bacterial isolates were round, the pathogenic isolate had a dark red center with white borders, while isolate C020 was yellow, isolate K007 with K009 was white and isolate K017 with K019 was clear white, and all isolates were classified as Gram negative. Based on the results of identification and characterization, it was found that the C020 isolate was a bacterium from the genus Erwinia sp. or Pantoea sp., while isolates K007, K009, K017 and K019 were bacteria from the genus Pseudomonas sp
