Abstract

Wortel merupakan salah satu komoditi sayuran yang memberikan kontribusi terhadap produksi hasil hortikultura nasional. Produksi wortel di Bali menunjukkan data yang bersifat fluktuatif dari tahun 2000 sampai 2018. Penurunan hasil wortel disebabkan oleh serangan OPT (Organisme Pengganggu Tanaman), diakibatkan oleh patogen Pectobacterium sp. penyebab busuk lunak pada umbi wortel. Upaya untuk mengendalikan penyakit adalah dengan menggunakan metabolit sekunder bakteri antagonis bakteri indigenous UB Forest. Penelitian bertujuan mengetahui potensi antagonis bakteri indigenous UB Forest terhadap patogen Pectobacterium sp. dan potensi kemampuan antagonisme bakteri indigenous UB Forest dan metabolit sekundernya dalam menghambat Pectobacterium sp. di cawan Petri dan menghambat penyakit busuk lunak pada umbi wortel. Penelitian dilaksanakan di Laboratorium Penyakit Tumbuhan 1 dan Laboratorium Toksikologi Jurusan Hama dan penyakit Tumbuhan Fakultas Pertanian Universitas Brawijaya, dimulai Bulan Desember 2019 sampai Maret 2020. Tahap pelaksanaan penelitian dilakukan mulai dari peremajaan bakteri Pectobacterium sp. dan bakteri Indigenous UB Forest, Uji patogenitas Pectobacterium sp. pada wortel, uji seleksi antagonis bakteri Indigenous UB Forest secara in Vitro, pembuatan ekstrak metabolit sekunder bakteri indigenous UB Forest, Uji antagonis metabolit sekunder terhadap patogen Pectobacterium sp. secara in Vitro, uji penghambatan metabolit sekunder bakteri indigenous UB Forest pada wortel secara in Vivo. Seleksi antagonis bakteri indigenous UB Forest menggunakan 6 isolat bakteri dari penelitian sebelumnya yang selanjutnya terpilih 2 isolat bakteri yaitu Xanthomonas sp. (N25) yang memiliki kemampuan lebih tinggi dari kontrol bakterisida Streptomicyn sulfat 20% dan Bacillus sp. (N3) memiliki kemampuan setara dengan kontrol bakterisisda Streptomycin sulfat 20%, isolat Xanthomonas sp. (N25) memiliki suatu senyawa yaitu bakterocin sedangkan isolat Bacillus sp. (N3) memiliki senyawa anti mikroba yang dapat menghambat patogen. Pada pengujian antagonis metabolit sekunder menunjukkan bahwa metabolit sekunder isolat Bacillus sp. (N3) dan isolat Xanthomonas sp. (N25) dapat menghambat pertumbuhan patogen Pectobacterium sp. dimana kemampuan metabolit sekunder setara dengan kontrol bakterisida Streptomycin sulfat 20% di dalam cawan Petri. Pada isolat Xanthomonas sp. (N25) dapat mengeluarkan suatu senyawa metabolit sekunder yang bersifat bakterisidal sedangkan Bacillus sp. (N3) mengeluarkan senyawa metabolit sekunder yang bersifat anti mikroba. Pada pengujian in Vivo tidak berperngaruh pada massa busuk lunak wortel. Pengujian bakteri Bacillus sp. (N3) dan Xanthomonas (N25) dan metabolit sekundernya dapat menghambat pertumbuhan Pectobacterium sp. di cawan Petri namun tidak dapat menghambat gejala busuk lunak di umbi wortel

English Abstract

Carrots are a vegetable commodity that contributes to the production of national horticulture products. Carrot production in Bali shows fluctuating data from 2000 to 2018. The decrease in carrot yield is caused by OPT (Organisme Pengganggu Tumbuhan), caused by the pathogen Pectobacterium sp. causes of soft rot in carrot root. The effort to control the disease is by using secondary metabolites of the antagonist bacteria of UB Forest indigenous bacteria. The study aims to determine the potential antagonist of UB Forest's indigenous bacteria against the pathogen Pectobacterium sp. and the potential antagonism ability of UB Forest's indigenous bacteria and its secondary metabolites in inhibiting Pectobacterium sp. in the Petri dish and inhibit soft rot disease in carrot root. The research was conducted at the Plant Disease Laboratory 1 and the Toxicology Laboratory, Department of Pests and Plant diseases, Faculty of Agriculture, Universitas Brawijaya, starting in December 2019 until March 2020. The implementation phase of the research was carried out starting from the rejuvenation of the bacterium Pectobacterium sp. and UB Forest Indigenous bacteria, Pathogenicity of Pectobacterium sp. in carrots, Indigenous bacterial antagonist selection test in Vitro, making secondary metabolite extracts of UB Forest indigenous bacteria, Test for secondary metabolite antagonists against pathogen Pectobacterium sp. in Vitro, the inhibition of secondary metabolites of UB Forest indigenous bacteria in carrots in Vivo. UB Forest's indigenous bacterial antagonist selection used 6 bacterial isolates from previous studies, which subsequently selected 2 bacterial isolates namely Xanthomonas sp. (N25) which has a higher ability than the bactericidal control of Streptomicyn sulfate 20% and Bacillus sp. (N3) has an equivalent ability to bactericidal control of Streptomycyn sulfate 20% , isolate Bacillus sp. (N3) has anti-microbial compounds that can inhibit pathogens. The secondary metabolite antagonist showed that the secondary metabolite isolate Bacillus sp. (N3) and Xanthomonas sp. (N25) can inhibit the growth of pathogenic Pectobacterium sp. where the ability of secondary metabolites is equivalent to the bacterisidal control of the Streptomycin sulfate 20% in the Petri dish. In Xanthomonas sp. (N25) can release a secondary metabolite compound that is bactericidal while Bacillus sp. (N3) release secondary metabolites which are anti-microbial. In Vivo testing did not affect the carrot soft rot mass. Bacillus sp. (N3) and Xanthomonas (N25) and their secondary metabolites can inhibit the growth of Pectobacterium sp. in a Petri dish but cannot inhibit the symptoms of soft rot in carrot root.
