Abstract

Radiasi gamma sering digunakan dalam radioterapi, sinar gamma memiliki energi yang besar dibandingkan dengan radiasi elektromagnetik lainnya sehingga dapat menembus jaringan manusia cukup jauh dan dapat membunuh sel kanker, tetapi di sisi lain penyerapan energi radiasi ke dalam tubuh akan dapat menyebabkan timbulnya radikal bebas pada dalam tubuh. Tujuan dari penelitian ini adalah mengetahui pengaruh dari radiasi gamma terhadap gambaran mikroskopis dan kadar sgpt organ hati mencit dan pengaruh pemberian ekstrak daun sirsak dan kulit manggis. Penelitian ini dilakukan dengan cara mencit dipapari radiasi gamma tanpa pemberian ekstrak dengan 5 variasi waktu paparan untuk mendapatkan dosis paparan maksimumnya. Kemudian, mencit diberi ekstrak dengan variasi dosis dengan masing-masing diberi durasi paparan 40 menit dengan total dosis 268 μSv. Mencit kemudian diukur kadar SGPT dalam darah mencit selanjutnya mencit dibedah dan dibuat preparat organ hati. Kerusakan organ dapat dilihat dari gambaran mikroskopis dengan perbesaran 100x, hasil dari kadar SGPT turut memperkuat hasil gambaran mikroskopis. Hasil penelitian menunujukkan sebelum diberi antioksidan, kerusakan total sel hepatosit adalah 47,34%. Setelah diberi ekstrak daun sirsak kerusakan total sel hepatosit adalah 25,02%, ekstrak kulit manggis 23,42% dan campuran ekstrak daun sirsak dan kulit manggis 21,95%. Untuk kadar SGPT memiliki nilai masing- masing 85,3 U/L, 77,7 U/L dan 72,6 U/L dari kadar awal yaitu 128,67 U/L.

English Abstract

Gamma radiation is often used in radiotherapy, gamma rays have greater energy compared to other electromagnetic radiation, that it can penetrate human tissue deep enough and can kill cancer cells, but on the other hand absorbed of radiation energy into the body can cause free radicals to occur in the body. The purpose of this study was to determine the effect of gamma radiation on microscopic images and levels of mice liver organs and the effect of giving soursop leaf extract and mangosteen peel. This research was carried by on mice exposed to gamma radiation without administration of extracts with 5 variations in exposure time to obtain the maximum exposure dose. Then, mice were given extracts with variant doses with a duration of exposure of 40 minutes with a total dose of 268 μSv. mice then measured SGPT levels in the blood of mice and then the mice were dissected and the liver preparations were made. the sctuctural damage can be seen from microscopic images with 100x magnification, the SGPT level results also strengthen the results of microscopic images. The results showed that before being given antioxidant, total hepatocyte cell damage was 47.34%. After being given soursop leaf extract total damage to hepatocyte cells were 25.02%, mangosteen peel extract 23.42% and a mixture of soursop leaf extract and mangosteen peel 21.95%. SGPT levels were 85.3 U/L, 77.7 U/L and 72.6 U/L respectively from the initial level of 128.7 U/L.
