Abstract
Tujuan: Penelitian ini bertujuan untuk mengetahui efektifitas deksametason, mitomycin-c dan 5- fluorourasil terhadap ekspresi alpha smooth muscle actine (α-sma) jaringan socket kontraktur. Metode: Kultur fibroblas dalam penelitian ini diambil dari satu pasien dengan kontraktur soket dan menjalani operasi rekonstruksi soket. Kultur fibroblas dibagi menjadi 4 kelompok. Setiap kelompok kultur fibroblas mendapat paparan dari agen antifibrotik yang berbeda. Kelompok I menerima deksametason 16 mg / ml, kelompok II menerima mytomicin-c 0,4 mg / ml, kelompok III menerima 5- fluorourasil 50 mg / ml, sedangkan kelompok IV adalah kelompok kontrol dan tidak mendapat paparan apapun. Pemeriksaan α-sma dilakukan terhadap myofibroblas socket kontraktur, dan dilakukan pencatatan hasil dari 5 kali pembacaan. Kemudian dilakukan analisa statistik dari hasil pembacaan tersebut. Results: Ekspresi α-sma mengalami penurunan pada semua kelompok paparan dibandingkan kelompok kontrol, dengan rata-rata sel myofibroblas paling banyak pada kelompok kontrol (83,02), diikuti kelompok deksametason (63,10), kemudian kelompok 5-fluorouracil (37,00), dan paling sedikit pada kelompok mitomycin-c (30,00). Hasil analisa statistik didapatkan perbedaan efektifitas yang signifikan dari ekspresi dari α-sma kultur sel myofibroblas dari kelompok yang mendapatkan paparan deksametason, mytomicin-c dan 5-flourouracil terhadap kelompok kontrol. Sedangkan pada perbandingan rata–rata ekspresi α-sma antara kelompok mytomicin-c terhadap kelompok 5-fluorouracil tidak didapatkan hasil yang signifikan. Kesimpulan: Pada penelitian ini perbandingan rata–rata ekspresi α-sma kultur sel myofibroblas jaringan soket kontraktur antara kelompok mytomicin-c dengan kelompok 5-fluorouracil tidak didapatkan perbedaan hasil yang signifikan, meskipun penurunan jumlah rata-rata sel myofibroblas kelompok mytomicin-c lebih tinggi dibandingkan penurunan rata-rata sel myofibroblas kelompok 5-flourouracil.
English Abstract
Purpose: This study aims to determine the effectiveness of dexamethasone, mitomycin-c and 5- fluorouracil on the expression of alpha smooth muscle actine (α-sma) from human socket contracture tissue. Methods: Fibroblast culture in this study was taken from one patient who had socket contractures and underwent reconstructive socket surgery. Fibroblast culture is divided into 4 groups. Each fibroblast culture group received exposure from different antifibrotic agents. Group I received dexamethasone 16 mg / ml, group II received mytomicin-c 0.4 mg / ml, group III received 5-fluorouracil 50 mg / ml, while group IV received received no exposure (control). Examination of α-sma antibody is performed on the contracted socket tissue myofibroblasts, and the results of 5 readings are recorded. Then statistical analysis of these readings was performed. Results: The expression of α-sma antibody decreased in all exposure groups compared to the control group, with the highest myofibroblast cell counts in the control group (83.02), followed by the dexamethasone group (63.10), then 5- fluorouracil group (37.00), and at least in the mitomycin-c group (30.00). From statistical analysis showed a significant decreased of the expression α-sma antibody in the exposure groups than the control group. Whereas the comparison of the expression α-sma between the mytomicin-c group to the 5-fluorouracil group were no significant. Conclusion: The were not significant result from comparison the expression of α-sma myofibroblast cells between the mytomicin-c group and the 5-fluorouracil group, although the mean decrease of myofibroblast cells count from mytomicin-c group was higher than the 5-flourouracil group.
