Abstract

Latar Belakang: Stress oksidatif merupakan hal penting yang berperan pada kondisi fibrosis hati. Peningkatan radikal bebas dapat dideteksi dengan peningkatan Malondialdehid (MDA) yaitu produk akhir dari peroksidasi lemak dan Glutation Peroksidase (GPx) adalah enzim antioksidan yang berperan menjaga integritas jaringan dari kerusakan oksidatif. Kurkumin sebagai salah satu antioksidan, antiinflamasi dan antifibrotik yang dipercaya dapat memicu regresi fibrosis hati. Tujuan: Mengetahui peran pemberian kurkumin terhadap kadar MDA plasma dan aktivitas GPx plasma pada tikus fibrosis hati sebagai salah satu respon terapi pada kondisi fibrosis hati. Metode: Penelitian eksperimental pada tikus jantan strain wistar. Terdapat 8 kelompok, masing-masing terdiri dari 4 tikus. Kelompok perlakuan kurkukim dipapar Karbon tetraklorida (CCl4) 1cc/kgbb 2x perminggu intraperitoneal selama 9 minggu untuk membentuk fibrosis F3. Kelompok Negatif (KN) diinjeksi Normal Salin. Setelah diinjeksi CCl4, Kelompok regresi spontan (KK) diberikan plasebo dan Kelompok perlakuan kurkumin (KP2, KP5 dan KP9) diberikan kurkumin 200mg/kgbb 2, 5 dan 9 minggu. Dilakukan analisis kadar MDA, aktivitas GPx plasma menggunakan uji Mann-Whitney, uji t independen, uji pengaruh dan lama pemberian kurkumin dengan ANOVA. Hasil: Terdapat peningkatan kadar MDA plasma namun tidak signifikan secara statistik dan peningkatan signifikan aktivitas GPx plasma pada tikus model fibrosis.Pemberian kurkumin menurunkan kadar MDA plasma yang signifikans pada KP5 dan KP9 dan meningkatkan aktivitas GPx plasma yang signifikan pada KP5. Kesimpulan: Pemberian kurkumin menurunkan kadar MDA plasma dan meningkatkan aktivitas GPx yang siginifikan pada minggu ke-5. Lama pemberian kurkumin berkorelasi dengan penurunan MDA dan peningkatan aktivitas GPx plasma.

English Abstract

Background: Oxidative stress play an important role in the pathogenesis of liver fibrosis. Increased the free radical can detected by the increasing of Malondialdehide (MDA) is an end product of lipid peroxidation in cell membrane and Glutatione Peroksidase (GPx) is an endogenous antioxidant enzyme which maintaining tissue integrity from oxidative damage. Curcumin as an antioxidant, anti-inflammatory,antifibrotic is believed to be one of the new therapy that can lead liver fibrosis regression. Objective: To Determine the effect of curcumin against oxidative stress reaction in Rat model liver fibrosis as one of respon treatment. Metode: An Experimental study in male rats wistar strain. There are 8 groups, 4 rats for each. KP group was exposed to CCl4 1cc/kg intraperitoneally 2 times per weeks for 9 weeks to form F3 Fibrosis. KN Group was injected Normal Saline. Once injected CCl4 control group (KK) was given a placebo and intervention group (KP) was Given curcumin 200mg per Kg weight for 2,5, and 9 weeks. Analysis of MDA level, GPx acitivy of plasma is conducted by independent t-test, influence test, and the duration of administration curcumin by ANOVA.Result: MDA level of plasma was increased but not statistically significant and Gpx activity of plasma was increased significantly in rat of liver fibrosis.Curcumin administration significantly reduced MDA level in KP5 and KP9 and the activity of GPx significantly increased in KP5. Conclusion: Curcumin administration significantly decreased the level of Plasma MDA and significantly increased the activity of plasma Gpx after 5 weeks.There was a coreelation between the duration of curcumin administration and decreased of MDA level and Gpx activity of plasma in regression of liver fibrosis.
