Abstract

Tanaman cabai menjadi salah satu komoditas yang mampu memberikan kontribusi terhadap produksi hasil sayuran nasional. Dalam pengelolaannya, tanaman cabai memiliki risiko yang cukup tinggi akan serangan hama dan penyakit yang mampu menurunkan hasil produksi dan menyebabkan kegagalan panen. Kendala utama dalam budidaya cabai adalah adanya serangan patogen. Salah satu patogen yang menyerang yaitu Cucumber mosaic virus (CMV). Upaya pengendalian yang dilakukan adalah memanfaatkan potensi mikroorganisme sebagai agensia pengendali hayati yaitu Bacillus subtilis dan mikoriza. Penelitian bertujuan untuk mengetahui potensi dari masing-masing agens hayati dan kombinasi dari keduanya dalam menginduksi ketahanan tanaman cabai terhadap CMV serta menguji kandungan senyawa hasil induksi ketahanan tanaman cabai terhadap CMV. Penelitian dilaksanakan di rumah kawat dan Laboratorium Penyakit Tumbuhan Universitas Brawijaya, dimulai bulan April sampai dengan September 2021. Tahapan penelitian meliputi penyemaian benih cabai, peremajaan isolat bakteri Bacillus subtilis, uji konsentrasi bakteri Bacillus subtilis, analisis pupuk mikoriza, uji sumber inokulum Cucumber mosaic virus (CMV), aplikasi pupuk mikoriza dan suspensi Bacillus subtilis serta inokulasi CMV pada tanaman cabai secara mekanis, pemasangan ajir dan pemeliharaan tanaman. Penelitian dilakukan dengan menggunakan Rancangan Acak Kelompok (RAK) dengan 7 perlakuan 5 ulangan. Data dianalisis dengan sidik ragam (ANOVA) dan Duncan Multiple Range Test (DMRT) pada taraf 5%. Pemberian Bacillus subtilis dan pupuk mikoriza dalam bentuk tunggal maupun kombinasi mampu memperpanjang masa inkubasi dengan memperlambat kemunculan gejala Cucumber mosaic virus (CMV) pada tanaman cabai. Perlakuan Bacillus subtilis dan pupuk mikoriza dalam bentuk kombinasi mampu menekan perkembangan penyakit Cucumber mosaic virus (CMV) sebesar 56,07%, serta memiliki kemampuan terbaik terhadap tinggi tanaman dan jumlah daun tanaman cabai. Perlakuan Bacillus subtilis dan pupuk mikoriza dalam bentuk kombinasi yang diinokulasi CMV menghasilkan peningkatan konsentrasi asam salisilat tertinggi dibandingkan perlakuan lain.

English Abstract

Chili plants are one of the commodities that are able to contribute to national vegetable production. In its management, chili plants have a high enough risk of pests and diseases that can reduce production yields and cause crop failure. The main obstacle in chili cultivation is the presence of pathogens. One of the pathogens that attack is Cucumber mosaic virus (CMV). Control efforts are made by utilizing the potential of antagonistic microorganisms as biological control agents, namely Bacillus subtilis and mycorrhizae. The aim of this study was to determine the potential of each biological agent and the combination of the two in inducing resistance of chili plants to CMV and to examine the content of compounds produced by chili plants’ resistance to CMV induction. The research was carried out at the wirehouse and the Plant Disease Laboratory of Universitas Brawijaya, from April to September 2021. The research stages included sowing chili seeds, rejuvenating Bacillus subtilis bacterial isolates, testing the concentration of Bacillus subtilis bacteria, analytical of mycorrhizae, test source of Cucumber mosaic virus (CMV) inoculum, application of mycorrhizal fertilizer and Bacillus subtilis suspension as well as mechanical inoculation of CMV on chili plants, installation of stakes and plant maintenance. The study was conducted using randomized block design with 7 treatments and 5 repeats. Data were analyzed with variance (ANOVA) and Duncan Multiple Range Test (DMRT) with a significance level of 5%. The application of Bacillus subtilis and mycorrhizal fertilizers in a single form or combination was able to extend the incubation period by slowing down the appearance of Cucumber mosaic virus (CMV) symptoms on chili plants. The treatment of Bacillus subtilis and mycorrhizal fertilizer in a combined form was able to suppress the development of Cucumber mosaic virus (CMV) by 56.07% and had the best ability on plant height and the number of leaves on chili plants. Treatment of Bacillus subtilis and mycorrhizal fertilizer in the form of a combination inoculated with CMV resulted in the highest increase in the concentration of salicylic acid compared to other treatments.
