Abstract
Dislipidemia merupakan faktor risiko utama yang berkontribusi terhadap aterosklerosis. Salah satu teori mengenai aterosklerosis adalah adanya keterlibatan lipoprotein-associated phospholipase A2 (Lp-PLA2). Darapladib merupakan penghambat selektif Lp-PLA2 yang hingga saat ini masih banyak pro dan kontra mengenai efeknya terhadap penghambatan proses aterosklerosis. Penelitian ini bertujuan untuk mengetahui efek darapladib terhadap ekspresi Lp- PLA2 dan molekul adhesi intracelluar adhesion molecule-1 (ICAM-1) dan vascular cell adhesion molecule-1 (VCAM-1) jaringan aorta tikus Sprague dawley model dislipidemia. Penelitian ini merupakan penelitian eksperimental, in vivo post test only controlled group design, menggunakan 24 tikus Sprague dawley dibagi menjadi enam kelompok berdasarkan dua serial waktu (normal 8 minggu, dislipidemia 8 minggu, dislipidemia yang diberi darapladib 20mg/200gram 8 minggu, normal 16 minggu, dislipidemia 16 minggu, dislipidemia yang diberi darapladib 20mg/200gram 16 minggu). Parameter yang dianalisis dalam studi ini antara lain ekspresi Lp-PLA2, ICAM-1, VCAM-1 jaringan aorta yang diukur dengan metode immunofluorescence dan diamati dengan confocal laser scanning microscope. Darapladib 20mg/200mg mampu menurunkan ekspresi ICAM-1 dan VCAM-1 jaringan aorta pada 8 minggu dan 16 minggu dan mampu menurunkan ekspresi Lp-PLA2 jaringan aorta pada 8 minggu
English Abstract
Dyslipidemia has been established as major risk factor contribute to atherosclerosis. One of emerging theory contribute to atherosclerosis is involvement of lipoprotein-associated phospholipase A2 (Lp-PLA2). Darapladib as inhibitor Lp-PLA2 have pro and contra about it is effect in atherosclerosis inhibition. This study aimed to examine the effects of darapladib on Lp-PLA2, intracellular adhesion molecule-1 (ICAM-1) dan vascular cell adhesion molecule-1 (VCAM-1) expression in aortic tissue in vivo. This was a true experimental laboratory, in vivo post test only control group design, with 24 Rattus novergicus Sprague dawley strain rats were divide into six group based on two serial time (normal group 8 weeks, dyslipidemic group 8 weeks, dyslipidemic group treated with darapladib 20mg/200 g b.wt 8 weeks and normal group 16 weeks, dyslipidemic griup 16 weeks, dyslipidemic group treated with darapladib 20mg/200 g b.wt 16 weeks). The parameter of this study were Lp- PLA2, ICAM-1, and VCAM-1 measured by immunofluorescence and observed with a confocal laser scanning microscope in aortic tissue. One way analysis of variance test showed Darapladib had significant effect (p<0.05) in decreasing ICAM-1 and VCAM-1 expression in 8 weeks and 16 weeks and Lp-PLA2 on 8 weeks in aortic tissue of hypercholesterol diet given Sprague dawley rat. Darapladib 20mg/200 g b.wt is proven to decrease ICAM-1 and VCAM-1 expression in aortic tissue in vivo. These finding provides a comprehensive mechanism inhibition of early progression of atherosclerosis through inhibition of adhesion molecules by darapladib
