Abstract

Studi farmakologi kontemporer telah menginformasikan bahwa Djulis (Chenopodium formosanum) dapat memberikan spektrum yang luas dari bioaktivitas, termasuk anti-adipogenesis, pemulihan cedera hati, dan antitumor. Djulis mengandung senyawa seperti polifenol, flavonoid, resveratrol, rutin asam, asam klorogenat, betanin dan pigmen isobetanin. Penelitian ini bertujuan untuk mengetahui pengaruh redaman ekstrak etanol Djulis hull (EEDH) pada kerusakan lambung yang diinduksi indometasin pada tikus. Aktivitas gastroprotektif EEDH dievaluasi menggunakan model tukak lambung yang diinduksi indometasin pada tikus setelah tiga minggu pengobatan. Tikus diberi 120 mg/kg indometasin dalam dua jam setelah yang terakhir hari pemberian oral 200 dan 500 mg/kg EEDH. Histopatologi penilaian dilakukan setelah dikorbankan. Indeks pencegahan ulkus, anti- enzim oksidatif dan ekspresi protein inflamasi dianalisis. Hasil penelitian menunjukkan bahwa pemberian oral 120 mg/kg indometasin secara signifikan meningkatkan area ulkus setinggi 2,65 ± 0,91 mm2 dan pretreatment dengan EEDH secara signifikan meringankan mukosa lambung kerusakan. 500 mg/kg EEDH menunjukkan efek yang baik pada redaman kerusakan dengan area ulkus terkecil dengan indeks pencegahan tertinggi setinggi 80,06%. 500 mg/kg EEDH secara signifikan meningkatkan aktivitas katalase (CAT) dan ekspresi COX-1, sedangkan ekspresi TNF-α adalah menurun. Oleh karena itu, EEDH mungkin memiliki potensi untuk pengembangan sebagai makanan fungsional untuk perlindungan lambung

English Abstract

Contemporary pharmacological studies have informed that Djulis (Chenopodium formosanum) may provide a broad spectrum of bioactivities, including anti-adipogenesis, recovery of liver injury, and antitumor. Djulis contains the compounds such as polyphenols, flavonoids, resveratrol, routine acids, chlorogenic acid, betanin and isobetanin pigments. This study aimed to explore the attenuation effect of ethanolic extract of Djulis hull (EEDH) on indomethacin-induced gastric damage in mice. The gastroprotective activity of EEDH was evaluated using the indomethacin-induced gastric-ulcer model in mice after three weeks treatment. The mice were gave 120 mg/kg of indomethacin in two hours after on the last day of 200 and 500 mg/kg EEDH oral administration. Histopathological assessment was performed after sacrificed. The ulcer preventive index, anti- oxidative enzyme and inflammatory protein expression were analyzed. The results showed that the oral administration of 120 mg/kg indomethacin significantly increased the ulcer area as high as 2.65±0.91 mm2 and pretreatment with EEDH significantly alleviated the gastric mucosa damage. 500 mg/kg of EEDH showed the good effect on attenuation of the damage with smallest ulcer area with the highest preventive index as high as 80.06 %. 500 mg/kg of EEDH significantly increased the activity of catalase (CAT) and the expression of COX-1, while the expressions of TNF-α were decreased. Therefore, EEDH might possess the potential for the development as a functional food for gastric protection

近代藥理學研究表明,台灣藜(Djulis)可能提供廣泛的生物活 性,包括減少脂肪的形成、肝損傷的恢復和抗腫瘤。其中台灣藜含有許 多化合物,例如多酚,類黃酮,白藜蘆醇,常規酸,綠原酸,甜菜鹼和 異丁烷素顏料。 這項研究主要目的是在探討台灣藜殼(EEDH)的乙醇提取物對吲 哚美辛誘導的小鼠胃黏膜損傷的保護作用。 在進行治療三週後,用吲哚美辛誘導的小鼠胃潰瘍模型評價 EEDH 對胃的保護作用。在分別給予 200 和 500 mg/kg EEDH 後的兩個小時或最 後一天,給小鼠服用吲哚美辛 120 mg/kg。處死後進行組織病理學評估。 分析潰瘍預防指數,抗氧化酶和發炎性蛋白的表達。 結果表明,口服 120 mg/kg 吲哚美辛可顯著增加潰瘍面積,達到 2.65±0.91 mm2,而 EEDH 預處理可顯著減輕胃黏膜損傷。 EEDH 500 mg/kg 對潰瘍的損害減輕效果最好,潰瘍面積最小,預防指數最高,高 達 80.06%。 EEDH 500 mg/kg 也顯著增加過氧化氫酶(CAT)的活性和 COX-1 的表達,而 TNF-α 的表達則降低。因此,EEDH 可能具有發展為 胃保護功能性食品的潛力
