Abstract

Transforming Growth Factor-β (TGF-β) merupakan factor pertumbuhan penting dalam proses penyembuhan luka. Kandungan Flavonoid dalam kulit jeruk nipis dapat menginduksi produksi TGF- β. Dalam membuat produk berbasis bahan alam dapat dilakukan pembentukan nanotransfersome untuk meningkatkan bioavailabilitasnya. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian nanotransfersome ekstrak kulit jeruk nipis (Citrus aurantifolia Swingle) terhadap ekspresi TGF-β oleh makrofag dalam penyembuhan luka mukosa labial tikus wistar. Metode penelitian true experimental in vivo dengan induksi termal mukosa labial tikus wistar, dibagi menjadi 4 perlakuan pada hari ke-3 dan ke- 7 dengan kelompok kontrol negatif (K-), kelompok kontrol positif dengan triamcinolone acetonide 0,1% (K+), Kelompok perlakuan ekstrak kulit jeruk nipis (PA), kelompok perlakuan nanotransfersome ekstrak kulit jeruk nipis (PB). Dilakukan pengecatan Imunohistokimia dan pengamatan dengan perbesaran 1000x untuk mengetahui ekspresi TGF-β oleh makrofag. Dari hasil uji One-way ANOVA, rata-rata jumlah ekspresi TGF-β tertinggi pada kelompok nanotransfersome. Sehingga dapat disimpulkan, pemberian nanotransfersome ekstrak kulit jeruk nipis berpengaruh terhadap peningkatan jumlah ekspresi TGF-β pada proses penyembuhan luka.

English Abstract

Transforming Growth Factor-β (TGF-β) is an important growth factor in wound healing process. The flavonoid substance in citrus peel believed could induce TGF-β. In making substance based on natural ingridients, nanoparticles can be formed to increase their bioavailability. The aim of this study was to determine the effect of nanotransfersome of Citrus Peel Extract (Citrus aurantifolia Swingle) Towards TGF-β Expression by macrophage in Wound Healing of White Rat’s Labial Mucosa. The experimental study in vivo had been carried out with the thermal induction of white rat’s labial mucosa which were divided into four groups. Then each group was induced on the 3rd and 7th day. The groups consist of : negative control group (K), positive control group with triamcinolone acetonide 0,1% (K+7), extract group (PA), nanotransfersome group (PB). Observation of TGF-β expression was carried out by immunohistochemistry staining process and light microscopy with magnification of 1000x. The result of data analysis using the one way ANOVA showed the highest average number of TGF-β expression in the nanotransfersome group. Thus, it can be concluded that nanotransfersome of Citrus Peel Extract (Citrus aurantifolia Swingle) could affected in increasing the number of TGF-β expression during wound healing in white rat’s labial mucosa.
