Abstract

-Sapi peranakan ongole (PO) merupakan plasma nutfah bangsa sapi potong yang memiliki keunggulan yaitu kemampuan beradaptasi yang mumpuni pada iklim tropis, memiliki daya cerna yang baik terhadap pakan dengan kandungan serat tinggi, serta tahan terhadap ekto dan endo parasit sehingga berpotensi untuk dikembangkan di seluruh Indonesia. Pengembangan populasi PO dapat dilakukan dengan teknologi IB yang didukung dengan produksi semen beku sapi PO yang berkualitas. Akan tetapi, proses pembekuan dapat menurunkan kualitas semen beku akibat dari produksi Reactive Oxygen Species (ROS) berlebih. Oleh karena itu, penambahan antioksidan seperti genestein dinilai mampu mengurangi produksi ROS berlebih sehingga dapat mempertahankan kualitas semen beku sapi PO. Penelitian ini bertujuan untuk mengetahui pengaruh lama ekuilibrasi suhu dingin dan ekuilibrasi uap nitrogen cair

English Abstract

The aim of this study is to determine the effect of different cold temperature equilibration and liquid nitrogen vapor equilibration time on the quality of ongole bull frozen semen supplemented with genistein. This study was held in UPT PT HMT Karangwaru Tuban from March until April 2021. This study method was a laboratory experiment that using nine combinations of cold equilibration and liquid nitrogen vapor equilibration time treatments. Semen was collected four times in a month period time from 7 years old ongole bull ear-tagged 0045, then one of ejaculated semen diluted using tris-aminomethane egg yolk-based extender and supplemented with genistein. Observed parameters are individual motility, viability, abnormality, and membrane integrity of spermatozoa after thawing. The result was statistically analyzed using a completely factorial randomized design and one-way analysis of variance (ANOVA). Post thawing observations show that different cold equilibration time has a significant (p<0.05) effect on sperm individual motility, abnormality, and membrane integrity. However, liquid nitrogen vapor equilibration time has a significant (p<0.05) effect on sperm individual motility and viability. Two hours cold equilibration time has the best result on maintaining sperm individual motility, abnormality, and membrane integrity respectively for 37.67 ± 2.58%, 11.14 ± 1.01%, and 48.95 ± 1.75% while ten minutes in liquid nitrogen vapor equilibration time has the best result on maintaining sperm individual motility and viability respectively for 37.00 ± 3.16% and 57.61 ± 2.04%.
