Abstract
Fibrosis hati merupakan tanda sirosis hati secara histopatologis. Karena sel utama pembentuk fibrosis hati adalah sel stelata hati (aHSC, activated hepatic stellate cells), apoptosisnya menjadi penting sebagai target utama terapi sirosis hati. TIMP-1 merupakan glikoprotein yang menghambat laju apoptosis melalui inhibisi protein MMP-2/3/7/9. Hal tersebut dapat berarti bahwa protein tersebut dapat meningkatkan fibrosis hati melalui inhibisi apoptosis aHSC. Telah diketahui pula bahwa induksi CCl4 pada tikus dapat mengakibatkan fibrosis hati dan pada paparan 12 minggu dapat menyebabkan fibrosis dan sirosis mikronoduler, tetapi belum diketahui hubungan ekspresi TIMP-1 aHSC dengan fibrosis hati pada tikus model fibrosis hati. Tujuan penelitian ini adalah untuk menjawab pertanyaan tersebut. Kami melakukan uji imunofluoresensi dan histopatologis untuk mengetahui hubungan antara ekspresi TIMP-1 aHSC dengan area fibrosis pada tikus galur Wistar Rattus novergicus terinduksi fibrosis hati (12 minggu paparan) (n = 5) dan tikus kontrol (n = 5). Didapatkan bahwa terdapat pengaruh signifikan antara ekspresi TIMP-1 aHSC dan area fibrosis.
English Abstract
Fibrosis is a histopathological sign of a liver cirrhosis. For HSCs are the main cellular cause of fibrosis, the apoptosis of these cells becomes significant in delivering cirrhosis treatment strategy. TIMP-1 is a glycoprotein attenuating the apoptosis through MMP-2/3/7/9 inhibition. Thus, can be concluded that the protein can induce fibrosis through aHSC apoptosis inhibition. For it is known that the mice model of CCl4 induction exhibits fibrosis’ as well as advanced micronodular cirrhosis for 12 weeks treatment, the question whether there are some correlation between TIMP-1 aHSC expression and fibrosis in mouse model still remains. The aim of the study was to answer the question. We performed immunofluorescence and histological assessment identifying the correlation between TIMP-1 aHSC expression and fibrosis area within Wistar strain rats Rattus novergicus (n=10) CCl4- induced (CCl4(+)) (n=5) and normal rats (CCl4(-)) (n=5). Given the significant correlation between TIMP-1 aHSC and the fibrosis area.
