Abstract
Avian Pathogenic Escherichia coli (APEC) merupakan E.coli strain patogen penyebab kolibasilosis pada unggas. Prevalensi kolibasilosis ayam broiler akibat APEC lebih banyak menginvasi secara sistemik dan menimbulkan gejala klinis berupa lesu, merunduk, kerdil, kerontokan bulu, dan diare. Penelitian ini bertujuan untuk mengidentifikasi E.coli serta mengetahui gambaran histopatologi pada organ hepar dan jantung ayam broiler yang diduga terinfeksi APEC. Sampel yang digunakan dalam penelitian ini adalah hepar dan jantung ayam broiler yang mengalami perubahan makroskopis dari 4 peternakan di Kabupaten Malang dengan riwayat kolibasilosis. Sebanyak 8 sampel dari 40 hepar dan jantung ayam broiler mengalami perihepatitis dan pericarditis ditandai dengan adanya eksudat fibrinosa, serta perubahan warna menjadi pucat. Isolasi dilakukan dengan menumbuhkan E.coli dari swab hepar dan jantung pada media Nutrient Broth (NB), dan Eosin Methylene Blue Agar (EMBA). Identifikasi dilanjutkan dengan pewarnaan Gram, serta ditanam pada media Indole Motility, Methyl red, Voges Proskauer, Citrat (IMViC), Triple Sugar Iron Agar (TSIA), uji fermentasi karbohidrat (glukosa, laktosa, sukrosa dan manitol), uji katalase dan Sorbirol Mac Conkey Agar (SMCA). Preparat histopatologi dari organ hepar dan jantung diamati dan dianalisis dengan uji Chi-Square Fisher untuk mengetahui hubungan infeksi APEC terhadap lesi-lesi pada hepar dan jantung ayam broiler. Hasil penelitian ini menunjukkan bahwa 7 sampel hepar dan jantung terinfeksi APEC dengan hasil analisa uji Chi-Square Fisher menunjukkan tidak ada hubungan yang signifikan antara infeksi APEC terhadap lesi-lesi pada organ. Pada pemeriksaan histopatologi hepar yang terinfeksi APEC menunjukkan kerusakan sedang yang terlihat adanya kongesti dan infiltrasi sel radang multifokal di sekitar pembuluh darah. Pada pemeriksaan histopatologi jantung yang terinfeksi APEC menunjukkan kerusakan sedang yang terlihat adanya infiltrasi sel radang pada pericardium dan diantara otot-otot jantung. Berdasarkan hasil dari gambaran histopatologi serta analisa statistika yang dilakukan pada hepar dan jantung dapat disimpulkan bahwa infeksi APEC tidak memberikan perubahan patologi secara signifikan pada organ hepar dan jantung ayam broiler.
English Abstract
Avian Pathogenic Escherichia coli (APEC) is a pathogenic E.coli that causes colibacillosis in poultry. The prevalence of broiler colibacillosis due to APEC is more systemic and showed clinical symptoms such as lethargy, ducking, dwarfism, hair loss, and diarrhea. This study has aimed to identify E.coli and determine the histopathological of broiler’s liver and heart that suspected APEC infections. The samples used were broiler's liver and heart that changed in macroscopic from 4 farms in Malang Regency with a history of colibacillosis. 8 samples of 40 liver and heart samples showed macroscopic changes that had perihepatitis and pericarditis are characterized by fibrinous exudate, and swelling of liver and heart. Isolation was carried out by growing E. coli from liver and heart swab on isolated media which are Nutrient Broth (NB) and Eosin Methylene Blue Agar (EMBA). Identification was continued with Gram staining and cultured on Indole Motility, Methyl red, Voges Proskauer, Citrate (IMViC) Triple Sugar Iron Agar (TSIA), carbohydrate fermentation test (glucose, lactose, sucrose, mannitol), catalase test and Sorbirol Mac Conkey Agar (SMCA). Histopathological of the liver and heart were observed and analyzed with Chi-Square Fisher test to determine the relationship of APEC infection to lesions in broiler’s liver and heart. The results of this study indicated that 7 liver and heart samples were infected with APEC and the results of Chi-Square Fisher test showed that there was no significant association between the appearance of APEC and lesions in the organs. Histopathologic examination of broiler’s liver that infected with APEC showed moderate damage, which showed congestion and mutifocal inflammatory cell infiltration around the blood vessels. Histopathologic examination of broiler’s heart that infected with APEC showed moderate damage, which was seen by inflammatory cell infiltration in the pericardium and between the heart muscles. Based on the results of histopathological and statistical analysis on liver and heart can be concluded that APEC infection doesn’t provide significant pathological changes in the broiler’s liver and heart.
