Abstract

English Abstract

Cancer is one of the most feared diseases, because the treatment process is very expensive. This disease develops rapidly and uncontrollably through cells in body tissues that are not normal. Factors from cancer are radiation, viruses, infections, cigarettes and chemicals from food. One of the alternative treatments used to cure cancer is to use plants in the environment. Plants that are widely used in cancer healing therapy are mangroves. In this study using mangroves with Avicennia marina species. The content of mangrove plants includes bioactive compounds including alkaloids, tannins, flavonoids, saponins and terpenoids, proven to have anticancer activity. The purpose of this study was to determine the IC50 value derived from Avicennia marina leaf extract. The method used in this research is the experimental method. The test parameters include yield, water content test, phytochemical test, toxicity test, total phenol test, cytotoxicity test and LC-MS test. The experimental design used was a simple completely randomized design (CRD) and was followed by the Tukey test. The results showed that the yield of n-hexane extract was 3.96%, ethyl acetate extract was 6.40% and ethanol extract was 14.40%. The results of the water content test in the n-hexane extract were 11.99%, the ethyl acetate extract was 12.80% and the ethanol extract was 16.57%. The results of the phytochemical test showed that the positive n-hexane extract contained alkaloids, steroids / triterpenoids and flavonoids. The positive ethyl acetate extract contains steroids / triterpenoids and tannins. The positive ethanol extract contains saponins, steroids / triterpenoids, flavonoids and tannins. Toxicity test using the BSLT method obtained LC50 yield values of 98.55 ppm in ethanol extract, 441.39 ppm in n-hexane extract and 1931.49 ppm in ethyl acetate extract. In the total phenol test results obtained a value of 113.22 mg / g GAE in the n-hexane extract, 752, 97 mg / g GAE in ethyl acetate extract and 1915.92 mg / g GAE in ethanol extract. Cytotoxicity testing on HeLa cells using the MTT assay method resulted in the highest viability value at a dose of 62.5 ppm of 56.800% and the lowest yield at a dose of 1000 ppm of 29.233%. For a dose of 500 ppm, it produces a viability value of 36.233%, a dose of 250 ppm of 42.733% and a dose of 125 ppm of 47.800%. The IC50 value generated from this study was 115.345 ppm which indicated that the ethanol extract of Avicennia marina leaves was quite active against HeLa cells. The results of LC-MS test showed that caffeine and diosmetin compounds were suspected. 800% and the lowest yield at a dose of 1000 ppm was 29.233%. The results of LC-MS testing showed the suspicion of caffeine and diosmetin compounds. 800% and the lowest yield at a dose of 1000 ppm was 29.233%. For a dose of 500 ppm, it produces a viability value of 36.233%, a dose of 250 ppm of 42.733% and a dose of 125 ppm of 47.800%. The IC50 value ix generated from this study was 115.345 ppm which indicated that the ethanol extract of Avicennia marina leaves was quite active against HeLa cells. The LC-MS test results showed the suspicion of caffeine and diosmetin compounds.
