Respon Imunologis Il1β Dan Il12 Ikan Kerapu Cantang (Epinephelus Sp.) Terinfeksi Viral Nervous Necrosis (Vnn) Dengan Pemberian Rekombinan Chlorella Vulgaris

Abstract

"Penggunaan vaksin rekombinan merupakan salah satu upaya untuk mengatasi penyakit pada ikan. Beberapa jenis vaksin VNN telah dikembangkan, termasuk vaksin yang dilemahkan formalin seperti vaksin subunit dengan protein lapisan rekombinan, vaksin partikel mirip virus, vaksin DNA dan vaksin rekombinan dari mikroalga laut Chlorella vulgaris. Tujuan dalam penelitian ini mengetahui status in-vivo pemberian vaksin rekombinan Chlorella vulgaris terhadap respon imunologis IL1Β dan IL12 Ikan Kerapu Cantang (Epinephelus sp.) yang terinfeksi Viral Nervous Necrosis (VNN). Metode peneitian yang digunakan dalam penelitian ini adalah metode eksperimen. Uji IL1β dan IL12 menggunakan uji sel antibody IgM pada ikan kerapu. Hasil penelitian respon IL1β menunjukkan titer ke 1 hingga titer tertinggi 1/2, dosis 33 μl (C) dan IL12 menunjukkan titer ke 1 perlakuan dosis 66 μl (G) dan 112 μl (H). Berdasarkan hasil penelitian yang telah dilakukan maka dapat diperoleh kesimpulan bahwa respon IL1β dan IL12 pada ikan kerapu cantang (Epinephelus sp.) sehat dengan perlakuan vaksin rekombinan Chlorella vulgaris direkomendasikan pada dosis 33μl (C) sedangkan pada perlakuan ikan penginfeksian VNN dengan perlakuan vaksin rekombinan Chlorella vulgaris direkomendasikan pada dosis 66 μl (G) dan 112 μl (H)."

English Abstract

The use of recombinant vaccines is one of the efforts to overcome diseases in fish. Several types of VNN vaccines have been developed, including formalin- attenuated vaccines such as subunit vaccines with recombinant coating proteins, virus-like particle vaccines, DNA vaccines and recombinant vaccines from the marine microalgae Chlorella vulgaris. Recombinant vaccines are the result of inserting viral genes into vectors that are expressed into competent bacterial cells. Vaccination in fish can be done by injection, immersion method and orally (mixed with feed). The purpose of this study was to determine the status of in vivo administration of recombinant Chlorella vulgaris vaccine against the immunological response of IL1β and IL12 Cantang Grouper (Epinephelus sp.) infected with Viral Nervous Necrosis (VNN). The research was conducted from February to June 2021 at UD. Giso Bangkit, Situbondo. The research method used in this research is the experimental method. Primary data consists of qualitative and quantitative data. The research design consisted of 8 treatments, namely (A) K(-): Healthy grouper without VNN infection, (B) K(+) : Healthy grouper with VNN infection, (C) C1: Healthy grouper without VNN infection with administration of 33 μl Chlorella vulgaris recombinant vaccine, (D) C2: Healthy grouper without VNN infection with 66 μl of recombinant Chlorella vulgaris vaccine, (E) C3: Healthy grouper without VNN infection with 112 μl of recombinant Chlorella vulgaris vaccine, (F) C4: Cantang grouper with VNN infection and administration of recombinant Chlorella vulgaris vaccine 33 μl, (G) C5: Grouper with VNN infection and administration of recombinant Chlorella vulgaris vaccine 66 μl (H) C6: Grouper with VNN infection and administration of recombinant vaccine Chlorella vulgaris 112 μl. Vaccine administration accompanied by coating using CFA on day 0 to day 4 was followed by giving IFA coating on the next day. The doses of recombinant vaccine coating concentrations for both CFA and IFA are 8.5 μl for treatment with recombinant Chlorella vulgaris vaccine 33 μl, 16.5 μl for treatment with recombinant Chlorella vulgaris vaccine 66 l, 28 l for recombinant vaccine Chlorella vulgaris 112 μl. Based on the study, the response of IL1β showed the 1st titer to the highest titer of 1/2, the 33 μl (C) and IL12 titers showed the 1st titer of treatment at 66 μl (G) and 112 μl (H). Based on the research that has been carried out, it can be concluded that the response of IL1β and IL12 in healthy cantang grouper (Epinephelus sp.) with recombinant Chlorella vulgaris treatment is recommended at a dose of 33μl, 33 μl (C) treatment, while in the treatment of VNN-infected fish with Chlorella vulgaris recombinant vaccine treatment is recommended at doses of 33 μl (F), 66 μl (G) and 112 μl (H). From this study, it is recommended that further research be conducted with 33 l of treatment, and 66 μl and 112 μl
