Abstract
Asma merupakan inflamasi kronik saluran napas, bersifat reversibel diakibatkan oleh berbagai faktor seperti alergen, infeksi saluran napas, cuaca dan lingkungan. Induksi hewan model asma dilakukan dengan pemberian ovalbumin (OVA) yang di perparah dengan lipopolisakarida (LPS) dari bakteri Phorphyromonas gingivalis. Kombinasi whey kefir dan tomat yang mengandung antioksidan seperti vitamin C berfungsi sebagai penangkal radikal bebas asma, probiotik sebagai imunomodulator dan senyawa peptida yang berperan sebagai antiinflamasi dan antimikroba. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian kombinasi whey kefir dan tomat terhadap ekspresi IL-4 dan histopatologi trachea tikus model asma. Metode penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan 5 kelompok perlakuan yaitu kelompok A (kelompok tikus sehat), kelompok B (positif asma) dan kelompok C,D,E (kelompok terapi). Ovalbumin diberikan sebanyak 3 kali pada hari ke 1,14 dan 21 dengan injeksi OVA I dan OVA II secara intraperitoneal dengan dosis 10μg/ml dan OVA III secara inhalasi dengan dosis 1 mg/ml selama 20 menit. Injeksi LPS secara intrasulkuler sebanyak 1 μg/ml diberikan pada hari ke 10 dan 11. Terapi kombinasi whey kefir dan tomat diberikan dengan dosis bertingkat 1 ml/ 200g BB, 1,5 ml/200g BB dan 2 ml/200g BB. Analisa kuantitatif ekspresi IL-4,rata-rata jumlah sel radang dan sel goblet trachea dilakukan dengan uji ANOVA dengan α=5% dan analisa kualitatif melalui pengamatan histopatologi trachea meliputi struktur susunan epitel, sel goblet dan sel radang menggunakan mikroskop pada perbesaran 400 x dan 1000x. Hasil penelitian didapatkan bahwa pemberian whey kefir dan tomat mampu menurunkan presentase IL-4, memperbaiki struktur susunan epitel trachea serta penurunan sel goblet dan sel radang pada tikus model asma. Dosis terapi terbaik yaitu 1,5 ml/200 g BB. Kesimpulan penelitian ini yaitu kombinasi whey kefir dan tomat dapat digunakan sebagai terapi tikus model asma.
English Abstract
Asthma is a chronic inflammatory in respiratory tract, reversible, caused by various factor such as allergens, infection of respiratory tract,weather and environment. Asthma is not only attack human, but animals too. Induction of animal model of asthma is using ovalbumin (OVA) and compunded with lipopolysaccharide (LPS) from bacteria Phorphyromonas gingivalis. The combination of tomato and whey kefir containing antioxidants such as vitamin C that have a function to against free radical, probiotic as imunomodulator and peptide compound that act as antiinflamation and antimicrobial. This research aims to know the effect of giving a combination of whey kefir and tomatoes toward the expression of IL-4 and histopathology of the trachea of rat as a animal model of asthma. The method on this research using randomized design with 5 group. Group A (healthy rat), Group B (asthma group) and Group C, D, E (therapy group). OVA were given 3 time on day 1, 14 and 21. OVA I and OVA II given by intraperitoneal injection with dose 10 μg/ml and OVA 3 is given by inhalation with dose 1 mg/ml. Injection of LPS did in intrasulculer as much as 1 μg/ml and given on day 10 and 11. Combination of whey kefir and tomatoes are given with dose 1 ml/200 g BW, 1.5 ml/200 g BW and 2 ml/200 g BW. Quantitative analyzed of IL-4 expression, inflammatory cell and goblet cell were analyzed using ANOVA test with α=5% and qualitative analyzed by observation of histopathology trachea include structure of trachea epithelium, goblet cell and inflammatory cell using microscope with 400x and 1000x magnification. The results showed that whey kefir and tomato were able to decrease IL-4 percentage, improve the structure of tracheal epithelial and decrease goblet cell and inflammatory cell in rats as animal model of asthma. The best therapy dose is 1.5 ml / 200 g BW. The conclusion of this research is the combination of whey kefir and tomato can be used as therapy in rats as animal model of asthma
