Abstract

Tujuan penelitian ini adalah menganalisis perubahan karakter morfologi dan kandungan kapsaisin, menginvestigasi variasi yang muncul, serta menganalisis polimorfisme DNA dan similaritas sepuluh tanaman cabai mutan G1/01/M4 hasil induksi mutasi dengan Ethyl Methanesulfonate (EMS) 0,01% generasi ke-4 dan kontrol berdasarkan karakteristik morfologi, kandungan kapsaisin dan profil molekuler. Deskripsi karakter morfologi mengacu pada deskriptor Capsicum spp. International Plant Genetic Resources Institute (IPGRI). Kandungan kapsaisin diukur menggunakan metode spektrofotometri, sedangkan profil genetik dianalisis secara molekuler RAPD. Analisis data statistika dilakukan menggunakan software SPSS 23, sedangkan matriks similaritas dibuat dengan software Multi-Variate Statistical Package Version 3.22. Hasil penelitian menunjukkan bahwa tanaman G1/01/M4 memiliki karakter yang lebih unggul dibandingkan kontrol yaitu lebar daun lebih besar, waktu pembungaan lebih cepat, diameter dan berat buah lebih besar, serta berat 100 biji kering lebih besar, sedangkan kekurangannya adalah jumlah buah atau produktivitasnya lebih sedikit dan tingkat kepedasan buah lebih rendah dibandingkan tanaman kontrol. Variasi masih tampak pada semua parameter yang diamati. Profil molekuler RAPD menghasilkan pita DNA polimorfik sebanyak 42 (78%) dan monomorfik sebanyak 14 (22%). Analisis similaritas berdasarkan karakter morfologi, kandungan kapsaisin dan profil molekuler menghasilkan lima kelompok. Kelompok I adalah T45, kelompok II yaitu T43 dan T40, kelompok III meliputi T29, T19, T18, T10 dan T8, kelompok IV meliputi T37dan T30 serta kelompok V yaitu tanaman kontrol. Tanaman G1/01/M4 yang paling berbeda dengan kontrol adalah T45.

English Abstract

The purpose of this study was to analyze the changes in morphological characters and capsaicin content, to investigate variations that arise, and to analyze DNA polymorphisms and similarity of ten G1/01/M4 mutant chili plants resulting from mutation with Ethyl Methanesulfonate (EMS) 0.01% the 4th generation and control based on morphological characteristics, capsaicin content and molecular profile. The capsaicin content was measured with spectrophotometric method, while the genetic profile was analyzed by molecular RAPD. Statistical data analysis was performed using SPSS 23 software while a similarity matrix was made with the Multi-Variate Statistical Package software Version 3.22. The results showed that the G1/01/M4 plant had a superior character compared control, which it has wider leaf width, faster flowering time, bigger diameter and weight of fruit, and greater weight of 100 dried seeds, while the inferior characters are lower average number of fruit and fruit spiciness. Variation is still visible in all parameters. The molecular profile of RAPD produced 42 (78%) polymorphic DNA bands and 14 (22%) monomorphics. Similarity analysis based on morphological characters, capsaicin content and molecular profile produced five groups. Group I is T45, group II are T43 and T40, group III are T29, T19, T18, T10 and T8, group IV are T37 and T30 and group V is control plant. The most different G1/01/M4 plant with control is T45.
