Abstract

Varietas anggur (Vitis vinifera) di Indonesia sangat beragam dengan karakteristik morfologi yang terlihat sama. Analisis kekerabatan dapat dilakukan berdasarkan penanda genetik seperti ISSR (Inter Simple Sequence Repeats). Tujuan penelitian ini adalah untuk mengetahui diversitas dan kekerabatan genetik 6 aksesi anggur menggunakan marka ISSR. Metode yang digunakan adalah deskriptif kualitatif dengan sampel 6 aksesi anggur. Data hasil penelitian diperoleh dari skoring pita DNA dan dianalisis menggunakan software NTSYS-pc berdasarkan UPGMA menggunakan metode SAHN. Sebanyak total 93 fragmen DNA, 74 (79.57%) fragmen DNA diantaranya merupakan polimorfik. Dendrogram hasil kekerabatan menunjukkan bahwa 6 aksesi anggur dikelompokkan menjadi dua kluster dengan koefisien similaritas sebesar 0.55-0.94. Tingkat koefisien similaritas tertinggi antara Jestro Ag 45 dan BS 80 (0.94) dan koefisien similaritas terendah antara BS 63 dengan Kediri Kuning/Jestro Ag 86 (0.55). Marka ISSR dapat digunakan untuk mengidentifikasi anggur dan dapat memberikan informasi yang berguna untuk program pemuliaan tanaman.

English Abstract

Grape varieties (Vitis vinifera) in Indonesia are very diverse with morphological characteristics that look the same. Analysis of genetic relationship can be done based on genetic markers such as ISSR (Inter Simple Sequence Repeats). The purpose of this study was to determine the diversity and genetic relationship of 6 grape accessions using ISSR markers. The method used in this study was descriptive qualitative with a sample of 6 grape accessions. The results of the research data were obtained from DNA band scoring and analyzed using NTSYS-pc software based on UPGMA using the SAHN method. A total of 93 DNA fragments, 74 (79.57%) of DNA fragments were polymorphic. The dendogram of genetic relationship showed 6 grapes accession were classified into two cluster with coefficient similarity 0.55-0.94. The highest coefficient similarity between Jestro Ag 45 and BS 80 (0.94), and the lowest between BS 63 with Kediri Kuning/Jestro Ag 86 (0.55). The ISSR marker can be used to indentify grape plants and can provide useful information for plant breeding programs.
