Abstract
Oviduct Spesifict Glikoprotein (OVGP) dikenalkan sebagai protein oviductal berpotensi mempengaruhi fertilisasi dan pembelahan embrio dini. OVGP diproduksi oleh epitel oviduktal non-silia pada banyak spesies mamalia. Pemberian OVGP pada spermatozoa sapi signifikan meningkatkan kapasitasi dan kemampuan untuk membuahi oosit sapi, serta meningkatkan motilitas dan viabilitas. Pemberian OVGP pada pengencer diyakini dapat berperan sebagai krioprotektan ekstraseluler spermatozoa sehingga mampu menurunkan abnormalitas spermatozoa dan ekspresi BCL-2. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dibagi menjadi 3 kelompok perlakuan, yaitu kelompok K sebagai kontrol (pengencer tidak ditambahkan OVGP) ; kelompok 1 (pengencer ditambahkan OVGP sebanyak 5 μl/ml) ; kelompok 2 (pengencer ditambahkan OVGP sebanyak 10 μl/ml). Pengukuran ekspresi BCL-2 dengan menggunakan imunoositokimia sedangkan untuk morfologi spermatozoa menggunakan pewarnaan eosin negrosin. Data didapatkan dengan melihat sel yang terekspresi dan menghitung sel mengekspresikan BCL-2 menggunakan software ImmunoRatio dan menghitung jumlah sel yang mengalami abnormalitas. Hasil penelitian menunjukkan ekspresi BCL-2 pada sspermatozoa terlihat pada bagian leher dan ekor spermatozoa. Terdapat penurunan rataan ekspresi BCL-2 pada perlkujan P1 dan P2 dibanding P0. Abnormalitas spermatozoa yang diamati yaitu no tail, no head, detached head, short tail, coiled tail, dag deffect, dan bent tail. Jumlah abnormalitas P1 dan P2 menurun dibanding P0. Berdasarkan hasil tersebut dapat disimpulkan bahwa terdapat penurunan ekspresi BCL-2 pada spermatozoa serta terdapat penurunan abnormalitas spermatozoa yang disuplementasi oleh OVGP pada pengencer semen kambing.
English Abstract
Oviduct Specific Glycoprotein (OVGP) is introduced as an oviductal protein that influences early fertilization and division of the embryo. OVGP is produced by non-ciliary oviductal epithelium in many mammalian species. Provision of OVGP in cow spermatozoa significantly increases capacitation and ability to fertilize bovine oocytes, as well as increase motility and viability. Giving OVGP to diluents can be done as spermatozoa extracellular cryoprotectants so as to reduce spermatozoa abnormalities and alter BCL-2. This study used a Completely Randomized Design (CRD) which was divided into 3 groups, namely the P0 group as a control (the diluent was not added OVGP); group P1 (diluent added OVGP by 5 μl / ml); group P2 (diluent added OVGP by 10 μl / ml). Measurement of BCL-2 expression using immunoocytochemistry while for spermatozoa morphology using negosin eosin staining. Data is obtained by looking at cells that are expressed and counting cells received by BCL-2 using the ImmunoRatio software and counting the number of cells that increase abnormality. The results showed the expression of BCL-2 in sspermatozoa was seen in the neck and tail of spermatozoa. It is estimated that the average decrease of BCL-2 in P1 and P2 requirements compared to P0. Spermatozoa abnormalities that are seen are no tail, no head, separate head, short tail, coiled tail, dag defects, and bent tails. The number of abnormalities P1 and P2 decreased compared to P0. Based on these results it can be concluded that there is a decrease in BCL-2 in spermatozoa and a decrease in abnormal spermatozoa supplemented by OVGP in goat semen diluents.
