Abstract

Gastritis merupakan istilah luas dari peradangan mukosa lambung. Penggunaan OAINS merupakan salah satu faktor risiko terbesar. Tanaman Kencana Ungu (Ruellia tuberosa L.) sudah lama dikenal sebagai tanaman obat. Daun tanaman ini banyak mengandung jenis flavonoid yang bermanfaat sebagai antiinflamasi. Penelitian ini dilakukan untuk melihat efek pemberian ekstrak daun Kencana Ungu terhadap penurunan kerusakan sel epitel lambung pada tikus yang diinduksi indometasin. Desain penelitian bersifat eksperimental dengan the Post Test Control Group Design. Penelitian menggunakan 5 kelompok sampel yaitu kelompok kontrol positif (ulkus diinduksi indometasin), kontrol negatif, serta kelompok yang diberi ekstrak daun Kencana Ungu dosis 100 mg/kgBB, 150 mg/kgBB, dan 200 mg/kgBB dengan pemberian indometasin sebagai inducer ulkus. Pembedahan dilakukan setelah perlakuan dan dipersiapkan untuk pengecatan Hematoxylin-Eosin. Kerusakan sel epitel lambung diamati secara histopatologis dengan skor Barthel-Manja. Analisa statistik didapatkan nilai signifikansi uji korelasi Pearson 0,000 dan correlation coefficient -0,877 yang menunjukkan nilai signifikan dan besar pemberian dosis ekstrak daun Kencana Ungu sebanding dengan penurunan lesi sel epitel lambung. Pemberian dosis 100 mg/kgBB dan 150 mg/kgBB menunjukkan hasil signifikan dibandingkan kontrol positif. Perlu digarisbawahi bahwasannya pemberian dosis 200 mg/kgBB menghasilkan nilai uji beda Multi Komparasi Metode Tukey sebesar 0,528 yang berarti telah mendekati kondisi kontrol negatif. Kesimpulan penelitian ini adalah pemberian ekstrak daun Kencana Ungu (Ruellia tuberosa L.) dengan dosis 100 mg/kgBB, 150 mg/kgBB, dan 200 mg/kgBB dapat menurunkan kerusakan sel epitel lambung pada tikus Rattus norvegicus galur Wistar yang diinduksi indometasin.

English Abstract

Gastritis is a broad term of inflammation of the gastric mucosa. The use of NSAIDs is one of the biggest risk factors. The Purple Kencana Plant ( Ruellia tuberosa L.) has long been known as a medicinal plant. The leaves of this plant contain many types of flavonoids that are useful as anti-inflammatory. This research was carried out to see the effect of giving Purple Kencana leaf extract to the reduction of gastric epithelial cell damage in indomethacin-induced mice. The research design is experimental with the Post Test Control Group Design . The study used 5 sample groups, namely the positive control group (indomethacin-induced ulcer), the negative control, and the group that was given Kencana Ungu leaf extract with a dose of 100 mg / kgBB, 150 mg / kgBW, and 200 mg / kgBW by giving indomethacin as inducer ulcer. Surgery is performed after treatment and prepared for Hematoxylin-Eosin staining. Gastric epithelial cell damage was observed histopathologically with Barthel-Manja scores. Statistical analysis obtained the significance value of the Pearson correlation test 0,000 and the correlation coefficient-0.877 which shows a significant value and large dose of Kencana Ungu leaf extract is proportional to the decrease in gastric epithelial cell lesions. Administering doses of 100 mg / kgBB and 150 mg / kgBB showed significant results compared to positive controls. It should be underlined that the administration of a dose of 200 mg / kgBB produces a Tukey Method Multi-Comparative test difference value of 0.528, which means it has approached a negative control condition. The conclusion of this study was that the administration of Kencana Ungu ( Ruellia tuberosa L.) leaf extract at doses of 100 mg / kgBB, 150 mg / kgBB, and 200 mg / kgBB can reduce gastric epithelial cell damage in Rattus norvegicus strains induced by Wistar strains.
