Abstract

LATAR BELAKANG:Ligamentum sakrouterina merupakan jaringan ikat penyokong organ panggul pada wanita. Pada Prolap Organ Panggul (POP) ligamentum sakrouterina sering mengalami kelemahan. Sel fibroblas sebagai salah satu penyusun utama jaringan ikat bertanggung jawab terhadap sintesis matriks ekstraseluler. Vitamin C sebagai salah satu kofaktor dalam penyusunan komponen matriks ekstraseluler (kolagen) berperan besar melalui jalur hidroksiprolin dan hidroksilisin sehingga terbentuk struktur triple helix kolagen dan cross link yang menstabilisasi kolagen.Sodium AscorbylPhosphatmerupakansenyawaturunandari vitamin C yang memilikisifatoksidasi yang lebihstabildibandingkan vitamin C. Penelitian paparan Sodium AscorbylPhosphat pada sel fibroblas yang dilakukan pada hewan coba atau manusia secara langsung terbukti meningkatkan proliferasi sel fibroblas, tetapi pengaruhnya terhadap sel fibroblas ligamentum sakrouterina wanita belum pernah dilakukan. Oleh karena itu perlu dilakukan penelitian mengenai pengaruh paparan Sodium AscorbylPhosphat terhadap proliferasisel fibroblas kultur ligamentum sakroutrina (in vitro) serta dosis yang adekuat yang disarankan untuk digunakan. TUJUAN:Tujuan penelitian iniadalahuntukmengetahui pengaruh paparan Sodium AscorbylPhosphat terhadap proliferasisel fibroblas kultur ligamentum sakrouterina pada wanita. METODE: Penelitian eksperimental sederhana non-bllinded dengan analisa pada akhir perlakuan (Post test control group design). Sampel dalam penelitian ini adalah seorang wanita dengan paritas minimal 2 dengan persalinan pervaginam yang menjalani operasi histerektomi. HASIL:Terdapat pengaruh signifikan paparan Sodium AscorbylPhosphatterhadap proliferasisel fibroblas kultur ligamentum sakrouterina pada dosis 400 μM, pada dosis 200μM mengalamipenurunandibandingkan control, sedangkanpadadosis 600 μM dan 800 μM tidak ada pengaruh signifikan. KESIMPULAN:Terdapat pengaruh signifikan paparan Sodium AscorbylPhosphat terhadap proliferasisel fibroblas kultur ligamentum sakrouterina, dalam kondisi in vitro dipengaruhi oleh proses kultur sel, proses pemaparan dan homeostasis.

English Abstract

BACKGROUND: The sacrouterine ligament is a connective tissue supporting the pelvic organs in women. On Pelvic Organ Prolapse (POP) the sacrouterine ligament often experiences weakness. Fibroblast cells as one of the main constituents of connective tissue are responsible for extracellular matrix synthesis. Vitamin C as one of the cofactors in the compilation of extracellular matrix components (collagen) plays a major role through the hydroxyproline and hydroxilisin pathways so that a collagen and cross link triple helix structure is formed which stabilizes collagen. Sodium Ascorbyl Phosphate is a derivative compound of vitamin C which has more stable oxidation properties than vitamin C. Research on exposure to Sodium Ascorbyl Phosphate on fibroblast cells carried out in experimental animals or humans is directly proven to increase fibroblast cell proliferation, but its effect on fibroblast cells sacrouterine ligaments women have never been done. Therefore it is necessary to do research on the effect of Sodium Ascorbyl Phosphate exposure on proliferation of sacroutrina ligamentous fibroblast cells (in vitro) and adequate doses which are recommended for use. OBJECTIVE: The purpose of this study was to determine the effect of Sodium Ascorbyl Phosphate exposure on proliferation of sacrouterine ligamentous fibroblast cells in women. METHOD: Simple non-blinded experimental study with analysis at the end of treatment (Post test control group design). The sample in this study was a woman with a minimum of 2 parity with vaginal delivery who underwent a hysterectomy operation. RESULTS: There was a significant effect of exposure to Sodium Ascorbyl Phosphate on proliferation of sacrouterine ligamentous fibroblast cells at a dose of 400 μM, at a dose of 200 μM it decreased compared to control, whereas there was no significant effect at 600 μM and 800 μM. CONCLUSION: There is a significant influence on Sodium Ascorbyl Phosphate exposure to proliferation of sacrouterine ligamentous fibroblast cells, in vitro conditions affected by cell culture processes, exposure processes and homeostasis.
