Abstract

Lupus Eritematosus Sistemik (LES) adalah penyakit autoimun sistemik yang disebabkan reaksi imun yang abnormal. Pengobatan LES saat ini yaitu kortikosteroid dan imunosupresan menimbulkan efek samping kerentanan terhadap infeksi. Metode lainnya yang sedang dikembangkan yaitu escalating dose self-antigen dsDNA immunotherapy. Tujuan penelitian ini adalah mengetahui pengaruh metode escalating dose self-antigen dsDNA immunotherapy pada LES terhadap kerentanan infeksi. Mencit Balb/c betina diinjeksi 0.5 ml pristane intraperitoneal. Sejumlah 24 mencit Balb/c betina dibagi menjadi 2 kelompok: 16 mencit diinjeksi 0.5 ml pristane intraperitoneal untuk menginduksi lupus dan 8 mencit dijadikan kontrol negatif. Dua belas minggu setelah injeksi, 8 mencit terinduksi lupus diberi dosis dsDNA 0,005 μg, 0,05 μg, dan 0,5 μg secara bertahap dengan selang waktu 1 minggu. dsDNA dikompleksikan dengan polyethylenimine (PEI). Ketiga kelompok mencit kemudian diberi sonde bakteri Salmonella Typhimurium 108 cfu/mL sebanyak 2 kali dengan selang waktu 3 hari. Tujuh hari setelah sonde kedua, dilakukan kultur bakteri pada organ usus halus seluruh mencit menggunakan media BSA. Escalating dose selfantigen dsDNA immunotherapy tidak memberikan pengaruh signifikan terhadap jumlah koloni Salmonella Typhimurium pada usus halus dibandingkan dengan kontrol positif (942.500 vs 78.250, p=0,081), namun masih terdapat perbedaan signifikan dibandingkan dengan kontrol negatif (942.500 vs 0, p=0,014). Dapat disimpulkan pemberian self-antigen dsDNA dosis 5x10-2μg, 5x10-1μg, 5μg secara bertahap tidak berpengaruh terhadap jumlah koloni Salmonella Typhimurium pada organ usus halus mencit Balb/c model lupus.

English Abstract

Systemic Lupus Erythematosus (SLE) is a systemic autoimmune disease caused by abnormal immune reactions. Current SLE treatments, namely corticosteroids and immunosuppressants, cause side effects of susceptibility to infections. Another method that is being developed is escalating dose self-antigen dsDNA immunotherapy. The purpose of this study was to determine the effect of the escalating dose self-antigen dsDNA immunotherapy method on susceptibility to infection in SLE. Female Balb/c mice were injected 0.5 ml intraperitoneal pristane. A total of 24 female Balb/c mice were divided into 2 groups: 16 mice were injected 0.5 ml pristane intraperitoneally to induce lupus and 8 mice were negative controls. Twelve weeks after injection, 8 lupus-induced mice were given an escalating dose of 0.005 μg, 0.05 μg, and 0.5 μg dsDNA with an interval of 1 week. dsDNA is complexed with polyethylenimine (PEI). The three groups of mice were then given oral administration of Salmonella Typhimurium 108 cfu/mL 2 times with an interval of 3 days. Seven days after the second administration, bacterial culture was carried out from the small intestine of all mice using BSA media. Escalating dose of self-antigen dsDNA immunotherapy had no significant effect on the number of Salmonella Typhimurium colonies in the small intestine compared with positive controls (942,500 vs 78,250, p = 0.081), but there were still significant differences compared with negative controls (942,500 vs 0, p = 0.014 ). It can be concluded that the administration of self-antigen dsDNA dose 5x10-2μg, 5x10-1μg, 5μg gradually has no effect on the number of Salmonella Typhimurium colonies in the small intestine of Balb/c mice lupus model.
