Abstract
Indometasin adalah salah satu NSAID yang dapat menimbulkan ulkus. Ulserasi akan terjadi di saluran cerna, terutama di lambung, dan usus halus. Dosis 30 mg/KgBB indometasin dapat menghasilkan senyawa radikal bebas akibat ulserasi dan kerusakan atau disfungsi mitokondria pada sel enterosit dan villus. Senyawa radikal bebas ini jika berikatan dengan lemak tak jenuh akan menghasilkan senyawa baru yaitu malondialdehida. Sehingga kadar senyawa radikal bebas dapat diukur melalui indikator kadar malondialdehida. Salah satu alternatif terapi stress oksidatif adalah dari tanaman tradisional yaitu daun kencana ungu yang diduga mempunyai aktivitas antioksidan sehingga kerusakan yang terjadi akibat senyawa radikal bebas dapat dihambat. 30 ekor tikus dibagi menjadi 5 kelompok, kelompok I kontrol negatif (K(-)) tanpa induksi indometasin, kelompok II kontrol positif (K(+)) indometasin 30 mg/Kg p.o, kelompok III perlakuan 1 (P(1)) indometasin 30 mg/Kg p.o + Ruellia tuberosa 100mg/kgBB, kelompok IV perlakuan 2 (P(2)) indometasin 30 mg/Kg p.o + Ruellia tuberosa 150mg/kgBB, kelompok V perlakuan 3 (P(3)) indometasin 30 mg/Kg p.o + Ruellia tuberosa 200mg/kgBB. Hasil penelitian ini ingin membuktikan induksi ekstrak Ruellia tuberosa dapat menurunkan kadar MDA jejunum secara signifikan dibandingkan dengan kontrol positif (p<0.05). Kesimpulan yang bisa diambil yaitu ekstrak daun kencana ungu dapat menurunkan kadar MDA jejunum tikus Wistar yang diinduksi indometasin.
English Abstract
Indomethacin is one NSAID that can cause ulcers. Ulceration will occur in the digestive tract, especially in the stomach and small intestine. 30 mg/KgBB doses of indomethacin can produce free radical compounds due to ulceration and damage or mitochondrial dysfunction in enterocyte and villus cells. These free radical compounds when bound to unsaturated fats will produce new compounds, namely malondialdehyde. So that the levels of free radical compounds can be measured through indicators of malondialdehyde levels. One alternative oxidative stress therapy is from traditional plants namely purple kencana leaves which are thought to have antioxidant activity so that damage caused by free radical compounds can be inhibited. 30 rats were divided into 5 groups, group I was negative control (K (-)) without indomethacin induction, group II was positive control (K (+)) indomethacin 30 mg / Kg po, group III treatment 1 (P (1)) indomethacin 30 mg / Kg po + Ruellia tuberosa 100mg / kgBB, group IV treatment 2 (P (2)) indomethacin 30 mg / Kg po + Ruellia tuberosa 150mg / kgBB, group V treatment 3 (P (3)) indomethacin 30 mg / Kg po + Ruellia tuberosa 200mg / kgBB. The results of this study want to prove the induction of Ruellia tuberosa extract can significantly reduce MDA jejunum levels compared with positive controls (p <0.05). The conclusion that can be drawn is the purple kencana leaf extract can reduce the MDA levels of indometacin-induced Wistar rats.
