Abstract

Menopause merupakan keadaan saat siklus menstruasi berhenti secara permanen disebabkan akibat ovarium yang gagal memproduksi ovum, yang terjadi minimal 12 bulan berturut-turut. Keadaan ini menyebabkan defisiensi hormon estrogen pada wanita. Hormon estrogen bermanfaat salah satunya dalam proses remodeling tulang. Daun Ciplukan (Physalis angulate L.) mengandung senyawa aktif physalin dan withanolide dimana menurut penelitian sebelumnya dapat berpotensi sebagai fitoestrogen. Selain itu terdapat kondisi hipoalbumin pada wanita menopause. Tujuan penelitian ini untuk mengetahui efektifitas pemberian ekstrak air daun ciplukan dan albumin berbagai dosis dalam menurunkan jumlah sel osteoklas pada tulang tibia tikus putih pasca ovariektomi. Metode yang digunakan adalah eksperimental, dimana penelitian ini menggunakan tikus model menopause atau tikus ovariektomi selama 5 minggu yang kemudian di terapi selama 4 minggu dengan ekstrak air daun physalis dikombinasikan dengan albumin berbagai dosis, yang kemudian dilakukan pembedahan untuk mengambil tulang tibia dan diamati perubahan jumlah sel osteoklas dengan mikroskop. Hasil penelitian menunjukan signifikansi saat diuji one-way ANOVA dengan nilai p = 0.001 dan p=0.000. Dan konsentrasi albumin saat ditambahkan ke ekstrak air daun ciplukan menunjukan korelasi negatif terhadap jumlah osteoklas pada tikus dengan nilai korelasi yang kuat (p = -0.659). Kesimpulan penelitian ini adalah ekstrak air daun Ciplukan dengan kombinasi albumin dapat menurunkan jumlah sel osteoklas secara in vivo.

English Abstract

Menopause is a condition when the menstrual cycle stops permanently due to an ovary that fails to produce the ovum, which occurs at least 12 consecutive months. This situation causes deficiency of the estrogen hormone in women. Estrogen is one of the main hormones of women, which highly functioning in regulation in the body, one of them is in the process of bone remodeling. Ciplukan (Physalis angulata L.) leaves contain active compounds of physalin and withanolide which according to previous studies can be potential as phytoestrogens. In addition there is a condition of hypoalbumin in menopausal women. The purpose of this study was to determine the effectiveness of administration of water extracts of ciplukan leaves and albumin in various doses in reducing the number of osteoclast cells in the tibia bones of post ovariectomized white rats. The method used was experimental, in which this study used a menopausal mouse model or ovariectomy mouse for 5 weeks which was then treated for 4 weeks with physalis leaf water extract combined with various doses of albumin, which after 9 weeks were then surgically removed to collect the tibia bone and observed changes in the number of cells osteoclasts under the microscope. The results showed significance when tested one-way ANOVA with p = 0.001 and p = 0.000. And the concentration of albumin when added to the water extract of ciplukan leaves showed a negative correlation with the number of osteoclasts in rats with a strong correlation value (p = -0.659). The conclusion of this study is the water extract of Ciplukan leaves with a combination of albumin can reduce the number of osteoclast cells in vivo.
