Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh zat pengatur tumbuh (ZPT) dan cahaya terhadap pertumbuhan dan komponen senyawa vetiver oil kultur kalus tanaman akar wangi (Vetiveria zizanioides L. Nash). Langkah penelitian yang dilakukan yaitu induksi kalus dari eksplan crown, kurva pertumbuhan kalus, subkultur kalus pada media perlakuan 2,4-D 0,5 mg/L dan kinetin 0,5 mg/L dibawah kondisi gelap dan terang. Analisis komponen senyawa vetiver oil menggunakan Gas chromatography mass spectrometry (GC-MS). Hasil penelitian menunjukkan bahwa kultur kalus dalam kondisi gelap menghasilkan indeks pertumbuhan, berat basah dan berat kering kalus lebih tinggi dibandingkan kondisi terang. Penambahan 2,4-D maupun kinetin pada konsentrasi 0,5 mg/L pada media kultur kalus tidak berpengaruh terhadap berat basah, berat kering dan indeks pertumbuhan. Berdasarkan kromatogram GC-MS, kultur kalus akar wangi teridentifikasi 58 komponen senyawa vetiver oil. Komponen utama senyawa vetiver oil pada kalus akar wangi yaitu kushimone (7,94%), vetiverol (6,58%), kushimene (6,44 %), β-vetivone (6,25%), α-vetivone (5,76%), prezizaene (2,09%) dan zizaene (1,04%). Cahaya dan ZPT dalam media berpengaruh terhadap komposisi dari beberapa komponen utama senyawa vetiver oil pada kultur kalus. Kondisi terang dan penambahan kinetin dalam media dapat meningkatkan komposisi senyawa vetiverol, β-vetivone dan α-vetivone, sedangkan kondisi gelap dan penambahan 2,4-D dapat meningkatkan komposisi senyawa kushimene, dan khusimone.

English Abstract

The prupose of this research were to determine the effect of plant growth regulator (PGR) and light on the growth and components of vetiver oil on callus culture of vetiver plants (Vetiveria zizanioides L. Nash). The research was conducted by several steps: callus induction from crown explants, callus growth curve, callus subculture on the treatment medium of 2.4-D 0.5 mg/L and kinetin 0.5 mg/L under conditions dark and light. Analysis vetiver oil components using Gas chromatography mass spectrometry (GC-MS). The results showed that callus culture under dark conditions produced the growth index, fresh weight and dry weight were higher than under light conditions. The supplemented with 2.4-D and kinetin at a concentration of 0.5 mg/L in callus culture media had no effect on fresh weight, dry weight and growth index. Based on the GC-MS chromatogram, vetiver callus were identified 58 components of vetiver oil compounds. Components of vetiver oil on callus vetiver were kushimone (7.94%), vetiverol (6.58%), kushimene (6.44 %), β-vetivone (6.25%), α-vetivone (5.76%), prezizaene (2.09%) and zizaene (1.04%). Light and PGR in the medium take effect the composition of several the main components of vetiver oil compounds in callus culture. Light conditions and the addition of kinetin in the media can increase the composition of the compound vetiverol, β-vetivone and α-vetivone, while dark conditions and the addition of 2,4-D can increase the composition of the compound kushimene, and khusimone
