Abstract
Penyakit hati kronis menurut Global Burden of Disease mortalitas karena sirosis hati mencapai 1 juta kematian pada tahun 2010. Adapun kerusakan sel hepatosit masih bisa regenerasi dan melakukan reaksi duktular yang ditandai dengan adanya cytokeratin 7 pada sitoplasma sel hepatosit. Reaksi duktular terjadi pada saat hati yang sudah mengalami kerusakan yang lebih parah dan hati tidak mampu untuk melakukan regenerasi hati. Pada hati yang mengalami fibrosis lebih parah, akan muncul adaptasi dari sel hepatosit dengan adanya vakuolisasi. Untuk mencegah progesivitas yang lebih parah bisa menggunakan Moringa oleifera (MO). MO dapat digunakan untuk menekan inflamasi dan menginduksi apoptosis dari sel yang rusak. Pada penelitian ini digunakan metode randomized posttest only controlled group yang bertujuan untuk mengetahui pengaruh pemberian ekstrak Moringa oleifera (MO) terhadap cytokeratin 7 (CK7) dan vakuolisasi sel hepatosit. Penelitian menggunakan 5 kelompok tikus dengan pembacaan menggunakan master Olyvia versi 2.4 DT for scan. Hasil uji Kruskal Wallis diketahui pemberian MO mencegah progesivitas kerusakan. Pemberian MO menunjukkan nilai yang signifikan pada dosis 600 mb/kgBB (p=0,000) dibandingkan dengan kelompok tanpa pemberian MO. akan searah dengan luas fibrosis, pada pemberian MO, vakuolisasi bertambah rendah dengan dosis MO yang tinggi yaitu pada dosis 600 mg/KgBB (p=0.00). Pada uji korelasi Spearman menunjukkan kekuatan korelasi sangat kuat, searah, dan signifikan antara dosis MO-CK7 (p=0.00; koefisien=0.798) dan kekuatan korelasi cukup, tidak searah, dan tidak signifikan antara dosis MO-(p=0.033; koefisien=0.864). Dapat disimpulkan bahwa pemberian MO berpengaruh terhadap penurunan kadar CK7.
English Abstract
Based on Global Burden of disease, hepatic cirhossis’s mortality reaches 1 million death in 2010. Hepatocyte could regenerate in acute damage. In chronic damaged liver, it would undergo ductular reaction and vacuolation would appear. Liver damage could be caused by viral and toxin such as carbon tetrachloride (CCl4). Research had shown that Moringa oleifera was effective to surpress inflammation reaction and to induce apoptose of the damaged hepatocyte. This study used randomized posttest only controlled group to understand the effect between Moringa oleifera (MO) extract on cytokeratin 7 (CK7) and cell vacuolation. In this study, 5 groups of mice were used and for interpretation used master Olyvia versi 2.4 DT for scan. Based on Kurskal Wallis test results, noted that MO administration prevented the progession of liver damage. The administration of MO showed a significant value at dose 600 mg/kgBW (p=0.000) compared to the group without MO administration. Cell vacuolization would align with the extent of fibrotic area. It would decrease eventually with MO administration at 600mg/kgBW dose (p=0.000). Spearman correlation showed a very strong, unidirectional and significant strenght between the MO dose – CK7 (p=0.000; coefficient=0.798) and on MO – cell vacuolation showed sufficient, not direct, and not significant correlation (p=0.033; coefficient= 0.864). It can be concluded that the administration of MO affects the decrease of CK7 level.
