Abstract

Diabetes neuropati merupakan komplikasi dari diabetes mellitus berupa gejala klinis atau subklinis yang mempengaruhi sistem saraf perifer. Pada diabetes, perubahan pada signalling ion kalsium berkontribusi terhadap perkembangan dari distal symmetrical sensorimotor polyneuropathy (DSN) dan nyeri. Peningkatan secara persisten pada Ca2+ sitosol juga terlibat dalam degenerasi saraf dan apoptosis sel. Penelitian ini bertujuan untuk menguji efek nimodipin terhadap ion kalsium (Ca2+) pada kultur cell line SH-SY5Y yang dipapar hiperglikemi kronik. Penelitian ini menggunakan model penelitian in vitro. Sebelum dilakukan pengukuran ion kalsium (Ca2+), sampel penelitian dibagi menjadi tiga kelompok paparan glukosa selama 6 hari, yaitu noromoglikemia 5 mM, hiperglikemia 25 mM, dan hiperglikemia 50 mM. Masing-masing kelompok diberi perlakuan tanpa nimodipin, nimodipin 10 nM dan nimodipin 100 nM selama 30 menit kemudian diperiksa parameter konsentrasi Ca2+ dengan pengecatan fluo-3 menggunakan confocal laser scanning microscopy (CLSM). Hasil penelitian menunjukkan bahwa induksi glukosa yang semakin tinggi mampu meningkatkan ekspresi ion kalsium secara signifikan. Pemberian nimodipin dosis 10 nM dan 100 nM mampu menurunkan ekspresi ion kalsium secara signifikan pada kelompok perlakuan glukosa 25 mM dan 50 mM. Namun pada kelompok glukosa 5 mM yang diberi nimodipin dosis 10 nM dan 100 nM justru meningkatkan ekspresi ion kalsium. Kesimpulan dari penelitian ini adalah pemberian nimodipin mampu menurunkan ekspresi ion kalsium pada sel SHSY5Y yang dipapar hiperglikemi kronik.

English Abstract

Diabetic neuropathy is a complication of diabetes mellitus which forms clinical or subclinical that affecting peripheral nervous system. In diabetes condition, changes in calcium ion signalling contribute to development of distal symmetrical sensorimotor polyneuropathy (DSN) and pain. Increasing of cytosolic calcium involved in neuron degenerative and cell apoptotic process. This study discuss the effect of nimodipin, a L-type calcium channel blocker, on calcium ions (Ca2+) in SH-SY5Y culture cell line exposed to chronic hyperglycemia. This study using in vitro research model. Before measuring calcium ions (Ca2+), the study sample was divided into three groups with 6 days of glucose administration; 5 mM noromoglycemia, 25 mM hyperglycemia, and 50 mM hyperglycemia. Each group were given different treatment ; without nimodipin, nimodipin 10 nM and nimodipin 100 nM for 30 minutes then proceed to obsevation of Ca2+ concentration parameters by fluo-3 dye using confocal laser scanning microscopy (CLSM). The results showed that higher dose of glucose could increase the calcium ion significantly. The administration of nimodipin at 10 nM and 100 nM doses significantly reduced calcium ions in the 25 mM and 50 mM glucose groups. In the 5 mM glucose group with administration of 10 nM and 100 nM nimodipin, Ca2+ have shown increased. The conclusion of this study is the administration of nimodipin could decreasing calcium ions in SH-SY5Y cells exposed to chronic hyperglycemia.
