Abstract

Tujuan penelitian ini untuk mengetahui pengaruh pupuk majemuk (NPK) terhadap keragaman, kerapatan kristal kalsium oksalat (CaOx), panjang idioblas kristal, dan jumlah anakan pada apu-apu (Pistia stratiotes). Penelitian ini menggunakan rancangan acak lengkap dengan 4 ulangan. Tahap awal penelitian, apu-apu diaklimasi selama 1 minggu dalam akuades, kemudian apu-apu dipindahkan ke wadah yang berisi air dan tanah seberat 4 kg dan diberi perlakuan pupuk NPK (15-15-15) P0 = 0 g, P1 = 0,2 g/kg, dan P2 = 0,4 g/kg. Pemupukan dilakukan pada 7 dan 30 HST dengan dosis masing-masing 50%. Pembuatan preparat diawali dengan pemotongan daun apu-apu ukuran 0,5 x 0,5 cm2. Potongan daun direndam dalam alkohol 100% (5 jam), NaOH 10% (1,5 jam), pemutih komersial 70% (2 menit), dan alkohol 30% (5 menit). Potongan daun di-mounting menggunakan gliserin dan di-seal menggunakan kuteks bening. Pengamatan kristal CaOx dan pengukuran idioblas kristal dilakukan menggunakan mikroskop binokuler Olympus CX 31 dan BX 53. Data yang diperoleh dianalisis ragam One-Way Anova (α = 0,05) menggunakan program SPSS Statistics 17.0. Pada daun apu-apu ditemukan dua bentuk kristal CaOx yaitu rafida dan druse. Kristal rafida yang ditemukan berbentuk berkas sejajar, tidak sejajar, dan saling tusuk. Pemberian pupuk NPK 0,4 g/kg memperendah kerapatan total kristal CaOx. Pemberian pupuk NPK sampai dosis 0,4 g/kg tidak berpengaruh terhadap ukuran idioblas kristal rafida dan druse. Namun, terlihat kecenderungan semakin tinggi dosis pemupukan, maka ukuran idioblas kristal rafida dan druse semakin pendek. Sebaliknya, jumlah anakan semakin banyak.

English Abstract

The purpose of this study was to determine the effect of compound fertilizers (NPK) on the diversity, density of calcium oxalate crystals, length of crystal idioblasts, and tillers number of Pistia stratiotes. This study used a complete randomized design with 4 replications. Initial step of method was acclimated P.stratiotes for 1 week in aquades. The plants were transferred to containers containing 4 kg of soil and water. They were treated with NPK fertilizer (15-15-15), P0 = 0 g, P1 = 0,2 g/kg, and P2 = 0,4 g/kg. Fertilization was given at 7 and 30 DAP with dose of 50% respectively. Leaf for microscopy observation was begun with cutting leaves 0,5 x 0,5 cm2. Pieces of the leaves were immersed in 100% alcohol for 5 hours, 10% NaOH for 1,5 hour, 70% commercial bleach for 2 minutes, and 30% alcohol for 5 minutes. Then the leaf pieces were mounted using glycerin and sealed using transparent nail polish. Observations and measurements of CaOx crystals were performed using an Olympus CX 31 and BX 53 binocular microscope. Obtained data were analyzed by One-Way Anova variance (α = 0.05) using the SPSS Statistics 17.0 program. The result showed that P. stratiotes have two forms of CaOx crystals namely raphide (bunch form) and druse. Raphide bunch crystals have various forms i.e. parallel, not parallel, and piercing each other. Fertilization (NPK) up to 0,4 g/kg lowered the total density of CaOx. Fertilization (NPK) up to a dose of 0,4 g/kg did not affect the size of raphide and druse crystal idioblasts. There was a tendency that the higher the dose of fertilizer gave the smaller the size of the raphide crystal and druse idioblast. In the other hand, numbers of tillers increased by higher fertilizer doses.
