Abstract

Aggregatibacter actinomycetemcomitans merupakan bakteri gram negatif penyebab penyakit periodontitis agresif. Daun ketapang (Terminalia catappa L.) diduga memiliki sifat antibakteri karena memiliki kandungan alkaloid, steroid, flavonoid, tanin, saponin, dan triterpenoid. Penelitian ini bertujuan untuk mengetahui efektivitas ekstrak daun ketapang (Terminalia catappa L.) dalam menghambat pertumbuhan bakteri Aggregatibacter actinomycetemcomitans secara in vitro. Penelitian ini merupakan eksperimental laboratorik true experimental post test only control group design yang menggunakan metode dilusi agar untuk mengukur kadar hambat minimum (KHM). Konsentrasi ekstrak yang digunakan 1%, 2%, 3%, 4%, 5%, 0% sebagai kontrol negatif, dan Chlorhexidine gluconate 0,2% sebagai kontrol positif. Hasil analisis data menggunakan Kruskal-Wallis dengan nilai signifikansi sebesar 0,000 (p<0,05) menunjukkan terdapat perbedaan signifikan terhadap kelompok perlakuan secara keseluruhan. Uji Mann-Whitney pada konsentrasi 1% dan 2% didapatkan nilai signifikansi sebesar 0,317 dan 0,127, karena nilai p>0,05 maka hasilnya tidak menunjukkan perbedaan signifikan dengan kontrol negatif, sedangkan nilai signifikansi konsentrasi 3%, 4%, 5% sebesar 0,008 (p<0,05) sehingga menunjukkan adanya perbedaan signifikan antara konsentrasi dengan kontrol negatif. Uji korelasi Spearman (r = -0,949 ; p<0,05) menunjukkan adanya korelasi negatif pada hubungan konsentrasi ekstrak dengan jumlah koloni. Kesimpulan penelitian ini adalah ekstrak daun ketapang (Terminalia catappa L.) efektif dalam menghambat pertumbuhan bakeri Aggregatibacter actinomycetemcomitans secara in vitro.

English Abstract

Aggregatibacter actinomycetemcomitans was a gram negative bacteria that causes aggressive periodontitis. Ketapang leaves (Terminalia catappa L.) contain antibacterial properties such as alkaloids, steroids, flavonoids, tannins, saponins, and triterpenoids. The aim of this study was to determine the effectiveness of ketapang leaf extract (Terminalia catappa L.) in inhibiting the growth of Aggregatibacter actinomycetemcomitans bacteria in vitro. This study was a true experimental laboratory experimental post test only control group design, that uses a dilution method to measure minimum inhibitory levels (MIC). The extract concentration used was 1%, 2%, 3%, 4%, 5%, 0% as negative controls, and Chlorhexidine gluconate 0.2% as a positive control. The results of Kruskal-Wallis test with a significance value of 0,000 (p <0.05) showed that there were significant differences in the overall treatment group. The Mann-Whitney test at a concentration of 1% and 2% obtained a significance value of 0.317 and 0.127, because the value of p> 0.05 then the results did not show a significant difference with negative controls, while the significance values of concentrations were 3%, 4%, 5% at 0.008 (p <0.05) which showed a significant difference between concentration and negative control. Spearman correlation test (r = -0.949; p <0.05) showed a negative correlation on the relationship of extract concentration with the number of colonies. The conclusion of this study was that ketapang leaf extract (Terminalia catappa L.) was effective in inhibiting the growth of Aggregatibacter actinomycetemcomitans bacteria in vitro.
