Abstract
Hama Plutella xylostella L. atau yang mempunyai nama umum hama ulat daun kubis adalah hama yang sangat penting pada tanaman kubis yang sulit untuk dikendalikan. Kerusakan dari serangan hama P. xylostella sangat berpengaruh dalam penurunan kuantitas dan kualitas dari hasil produksi tanaman kubis. Permasalahan resistensi hama P. xylostella yang tinggi , di timbulkan dari beberapa faktor yang mendukung. Faktor biologi yang terjadi pada hama merupakan kejadian yang sangat di pertanyakan. Maka perlu dilakukan eksplorasi serta penelitian lebih lanjut tentang potensi bakteri yang terdapat pada bagian pencernaan P. xylostella. Berbagai macam bakteri yang terdapat pada sistem percernaan khususnya bagian midgut (mesenteron), dapat bersimbiosis dan diduga dapat mendegradasi racun insektisida profenofos yang telah di aplikasikan. Penelitian dilaksanakan dengan pengambilan sampel P. xylostella L. di daerah Krajan, Pujon Kidul, Malang, Jawa Timur dan Laboratorium Bakteri Tumbuhan Jurusan Hama dan Penyakit Tumbuhan Fakultas Pertanian Universitas Brawijaya, Malang, Jawa Timur di mulai pada bulan November 2018 sampai Oktober 2019. Penelitian dilakukan dalam 5 tahap yaitu : (1) pengambilan sampel ulat Plutella xylostella (2) isolasi bakteri pada bagian midgut Plutella xylostella (3) seleksi bakteri pada bagian midgut Plutella xylostella (4) uji biodegradasi insektisida berbahan aktif profenofos (5) identifikasi bakteri terpilih sampai tingkat genus. Hasil isolasi dan seleksi kemampuan hidup pada media NA+profenofos di dapatkan 14 isolat bakteri yang mampu hidup. Uji hipersensitif di lakukan pada 14 isolat bakteri, didapatkan 10 isolat bakteri tidak berpotensi sebagai patogen dan 4 bakteri berpotensi sebagai patogen. Pada uji biodegradasi insektisida profenofos pada 14 isolat bakteri terpilih pada 3 konsentrasi insektisida yang berbeda yaitu : 0,75 ml/L, 1,5 ml/L dan 3 ml/L dan diamati selama 72 jam. Didapatkan 2 isolat yang mampu mendegradasi insektisida profenofos ditunjukkan dengan munculnya zona bening, 2 Isolat yang mampu mendegradasi insektisida antara lain isolat G8 dan isolat G9. Sedangkan untuk isolat G1, G2, G3, G4, G5, G6, G7, G10, G11, G12, G13 dan G14 hanya mampu tumbuh pada 3 konsentrasi insektisida namun tidak mampu mendegradasi insektisida tersebut. Hasil identifikasi 14 isolat bakteri yang didapatkan pada midgut P. xylostella antara lain : Pada isolat G2, G6, G7 dan G11 merupakan genus Erwinia sp., pada isolat G1, G3, G5, G8, G9 dan G12 merupakan genus Pantoea sp., sedangkan pada isolat G4, G10, G13 dan G14 merupakan genus Coryneform sp
English Abstract
Plutella xylostella L. or which has the common name of cabbage leaf caterpillar is a very important pest in cabbage plants that are difficult to control. Damage from P. xylostella pests is very influential in reducing the quantity and quality of cabbage production. The problem of high P. xylostella pest resistance, caused by several factors that support. Biological factors that occur in pests is a very questionable event. Then it is necessary to explore and further research about the potential of bacteria found in the digestive part of P. xylostella. Various kinds of bacteria found in the digestive system, especially the midgut (mesenteron), can be symbiotic and are thought to be able to degrade profenophos insecticide poison that has been applied. The research was carried out by taking P. xylostella L. samples in Krajan, Pujon Kidul, Malang, East Java and the Plant Bacteria Laboratory, Department of Pests and Plant Diseases, Faculty of Agriculture, Universitas Brawijaya, Malang, East Java, starting in November 2018 until October 2019. Research carried out in 5 stages, namely: (1) sampling of caterpillar Plutella xylostella (2) isolation of bacteria in the midgut section of Plutella xylostella (3) bacterial selection in the midgut section of Plutella xylostella (4) biodegradation test of insecticide with active ingredients from profenofos (5) identification of selected bacteria up to genus level. The results of isolation and selection of the ability to live on NA + profenofos media obtained 14 bacterial isolates that are able to live. Hypersensitivity test was carried out on 14 bacterial isolates, found 10 bacterial isolates had no potential as pathogens and 4 bacteria had potential as pathogens. In the biodegradation test of Profenophos insecticide on 14 selected bacterial isolates at 3 different insecticide concentrations namely: 0.75 ml / L, 1.5 ml / L and 3 ml / L and observed for 72 hours. Two isolates that were able to degrade profenophos insecticide were shown by the emergence of a clear zone, 2 isolates that were able to degrade insecticides include G8 isolates and G9 isolates. Whereas isolates G1, G2, G3, G4, G5, G6, G7, G10, G11, G12, G13 and G14 were only able to grow at 3 concentrations of insecticide but were unable to degrade the insecticide. The results of the identification of 14 bacterial isolates obtained in P. xylostella midgut include: In isolates G2, G6, G7 and G11 are the genus Erwinia, in isolates G1, G3, G5, G8, G9 and G12 are the genus Pantoea, whereas in isolates G4, G10, G13 and G14 are genus Coryneform.
