Abstract
Streptococcus mutans merupakan bakteri yang terdapat didalam rongga mulut yang berperan sebagai agen utama penyebab karies gigi, yang memiliki enzim glukosiltransferase (GTF). Enzim GTF akan mengubah sukrosa menjadi fruktosa dan glukosa. Lidah Buaya (Aloe vera L.) sebagai bahan alami memiliki sifat antibakteri. Dalam hal ini memiliki kandungan tanin yang secara spesifik dapat menghambat aktivitas enzim Glukosiltransferase Streptococcus mutans. Tujuan penelitian ini untuk mengetahui pengaruh ekstrak daun lidah buaya (Aloe vera L.) terhadap aktivitas enzim GTF Streptococcus mutans. Penelitian ini menggunakan ekstrak daun lidah buaya konsentrasi 12,5%, 25%, 50%, 75% dan 100% sebagai perlakuan, dan aquades sebagai kontrol. Enzim GTF diperoleh dari hasil sentrifugasi bakteri Streptococcus mutans. Aktivitas enzim GTF diukur dengan menghitung kadar fruktosa menggunakan High Performance Liquid Chromatography (HPLC). Hasil perhitungan aktivitas enzim GTF menunjukkan perbedaan penurunan kadar fruktosa yang signifikan antara kelompok perlakuan yang diberikan ekstrak daun lidah buaya dengan kelompok kontrol. Analisa data menggunakan Korelasi dan Regresi menunjukkan berpengaruh sebesar 76,1% terhadap penurunan kadar fruktosa. Berdasarkan penelitian daun lidah buaya dapat menghambat kerja dari enzim glukosiltransferase.
English Abstract
Streptococcus mutans is a bacteria in the oral cavity which consists of glucosyl transferase (GTF) enzyme and acts as the main agent that causes dental caries. GTF enzyme will convert sucrose into fructose and glucose. Aloe vera (Aloe vera L.) is one of herbal plants which has a antibacterial effect. Aloe vera has active substancse such as tannin which specifically can inhibit glucosyltransferase (GTF) enzyme Streptococcus mutans. The purpose of this research is to investigate the effect of Aloe vera leaf (Aloe vera L.) extract to the activity of Streptococcus mutans’s GTF enzyme. This research used 12,5%, 25%, 50%, 75% and 100% concentration of Aloe vera extract and count as the treatment group, and aquadest as a control group. The GTF enzyme is obtained from the supernatant of Streptococcus mutans. The activity of GTF enzyme is measured by calculating the fructose concentration using High Performance Liquid Chromatography (HPLC). The results of calculations of enzyme activity GTF shows the difference a significant fructose levels between treatment groups given the Aloe Vera leaf extract with the control group. Data analysis using correlation and regression shows the effect of 76.1% against a decline in the levels of fructose. Based on this research, it can be concluded that leaves of the Aloe Vera can inhibit the action of the glucosyltransferase enzyme.
