Abstract

Ulkus traumatik merupakan ulkus pada rongga mulut yang disebabkan oleh trauma. Sel makrofag pada penyembuhan luka berperan dalam fagositosis, sekresi sitokin dan growth factor. Gel ekstrak etanol biji kedelai mengandung isoflavon yang dapat meningkatkan aktivitas makrofag dan TGF- β. Tujuan penelitian ini adalah untuk mengetahui apakah gel ekstrak etanol biji kedelai (Glycine max (L.) Merill) berpengaruh terhadap jumlah makrofag pada penyembuhan ulkus traumatik mukosa labial tikus putih. Metode yang digunakan adalah true experimental menggunakan Randomized Post Test Only Controlled Group design secara in vivo. Penelitian ini dibagi menjadi 8 kelompok dengan 2 time series yaitu 2 dan 6 hari. Dosis gel ekstrak etanol biji kedelai yang digunakan yaitu 0%, 40%, 60%, dan 80%. Setelah pengorbanan, jaringan ulkus diambil dan dilakukan pewarnaan Hematoxylin Eosin, kemudian sel makrofag diamati menggunakan mikroskop digital. Hasil Post Hoc Tukey menunjukkan bahwa gel ekstrak etanol biji kedelai dengan konsentrasi 80% secara signifikan berpengaruh terhadap jumlah makrofag pada penyembuhan ulkus traumatik mukosa labial tikus putih dibandingkan kelompok lain pada hari ke-2 dan 6. Makrofag mencapai puncak pada hari ke-2 dan menurun pada hari ke-6. Berdasarkan hasil penelitian, dapat disimpulkan bahwa gel ekstrak etanol biji kedelai (Glycine max (L.) Merill) berpengaruh terhadap jumlah makrofag pada penyembuhan ulkus traumatik mukosa labial tikus putih dan konsentrasi 80% efektif untuk penyembuhan ulkus traumatik.

English Abstract

Traumatic ulcers are oral ulcers caused by trauma. Macrophage cells in wound healing play a role in phagocytosis, cytokine secretion and growth factor. The soybean seeds ethanol extract gel contains isoflavones which can increase macrophage activity and TGF-β. The aim of this study was to determine whether the soybean seeds (Glycine max (L.) Merill) ethanol extract gel had an effect on the number of macrophages in labial mucosa traumatic ulcer healing of white rats. The method used is true experimental using Randomized Post Test Only Controlled Group design in vivo. This study was divided into 8 groups with 2 time series, 2 and 6 days. The doses of ethanol extract of soybean seeds used were 0%, 40%, 60%, and 80%. After sacrifice, the ulcer tissue was taken and Hematoxylin Eosin staining was performed, then macrophage cells were observed using an digital microscope. The Post Hoc Tukey results showed that 80% soybean seeds ethanol extract gel with a significant effect on the number of macrophages in labial mucosa traumatic ulcer healing of white rats compared to other groups on days 2 and 6. Macrophages reach a peak on day 2 and decrease on day 6. Based on the results of the study, it can be concluded that the soybean seeds (Glycine max (L.) Merill) ethanol extract gel affects the number of macrophages in labial mucosa traumatic ulcer healing of white rats and an 80% concentration effective for healing traumatic ulcers.
