Abstract
Inflammatory Bowel Disease (IBD) adalah penyakit gastrointestinal kronis disebabkan karena faktor kecacatan permeabilitas, genetik, iskemik, biokimia dan kelainan prikosomatik, infeksi dan agen parasit, dan efek negatif obat-obatan NSAID salah satunya adalah Indometasin. Penelitian ini bertujuan untuk mengetahui perbedaan efektifitas ekstrak daun P. longifola yang berasal dari Indonesia dan Filipina terhadap ekspresi ERK dan gambaran histopatologi pada tikus model IBD. Penelitian ini menggunakan hewan coba tikus (Rattus norvegicus) jantan strain Wistar berumur 2-3 bulan dengan berat 150 – 200 gram. Hewan coba dibagi menjadi 3 kelompok yaitu kelompok terapi menggunakan goal standard (Sulfasalazine dosis 20 mg/kgBB) secara per oral, kelompok dengan menggunakan ekstrak P. longifolia Indonesia (dosis 300 mg/kg BB) secara per oral, dan kelompok menggunakan ekstrak P. longifolia Filipina (dosis 300 mg/kgBB) secara per oral selama 7 hari. Data dianalisis dengan ANOVA dan dilanjutkan dengan uji lanjutan BNJ (Beda Nyata Jujur) dengan α= 0,05%. Kemudian untuk melihat perbedaan antara terapi ekstrak Indonesia dan Filipina dianalisis dengan Independent T-Test. Hasil ekspresi ERK diamati dengan metode imunohistokimia dan perbaikan histopatologi mukosa kolon diamati dengan pewarnaan Hematoxylin-Eosin (HE). Hasil perhitungan ANOVA menunjukkan ekspresi ERK pada kelompok terapi Sulfasalazine kelompok terapi ekstrak etanol daun P. longifolia dari Indonesia, dan kelompok terapi ekstrak etanol P. longifolia dari Filipina tidak berbeda signifikan (P>0,05). Hasil uji Independent T-Test pada kelompok terapi ekstrak etanol daun P.longifolia dari Indonesia memiliki rata-rata ekspresi ERK lebih tinggi daripada kelompok terapi ekstrak etanol daun P.longifolia dari Filipina. Pada gambaran histopatologi kolon terlihat adanya perbaikan kolon dan berkurangnya infiltrasi sel radang.
English Abstract
Inflammatory Bowel Disease (IBD) is a chronic gastro intestinal disease caused by disability, genetic, ischemic, biochemical and procosomatic abnormalities, parasitic infections and agents, and the negative effects of NSAIDs, one of which is indomethacin. This study aims to determine the differences in effectiveness of P. longifolia leaf extract originating from Indonesia and the Philippines against ERK expression and histopathological features in IBD model mice. This study used experimental animals, rats (Rattus norvegicus), Wistar strain, aged 2-3 months, weighing 150-200 grams. The experimental animals were divided into 3 groups, namely the therapy group using a standard goal (Sulfasalazine dose of 20 mg / kgBW) orally, the group using P. longifolia Indonesia extract (dose 300 mg / kgBW) orally, and the group using extract of P. longifolia from Philippines (dose 300 mg / kgBW) orally for 7 days. Data were analyzed by ANOVA and continued with continued BNJ (Honestly Different Difference) test with α = 0.05%. Then to see the differences between Indonesian and Philippine extract therapies were analyzed by the Independent T-Test. The results of ERK expression were observed by immunohistochemistry and histopathological repair of colon mucosa was observed by Hematoxylin-Eosin (HE) staining. The results showed that ERK expression in therapy group of Sulfasalazine, therapy group of P. longifolia ethanol extract from Indonesia, and therapy group of P. longifolia ethanol extract from the Philippines are not significantly different. The results of the Independent T-Test in the P. longifolia ethanol extract therapy group from Indonesia had a higher ERK expression than the therapeutic group ethanol extract of P. longifolia from the Philippines. In the histopathology view of colon, there is an improvement in the colon and reduced inflammatory cell infiltration.
