Abstract
Infeksi bakteri pada luka kastrasi dapat memperlama kesembuhan luka. Luka kastrasi yang tidak aseptis dapat menjadi port of entry dari bakteri patogen Staphylococcus aureus sehingga luka menjadi semakin parah dan sembuh lebih lama. Oleh karena itu dibutuhkan terapi untuk mempercepat kesembuhan luka serta memiliki sifat antimikroba untuk mengatasi infeksi pada luka kastrasi. Penelitian ini bertujuan untuk mengetahui ekspresi TGF-β serta histopatologi pada jaringan luka kastrasi yang diterapi menggunakan gel getah pohon jaran (Lannea coromandelica). Gel getahpohon jaran dibuat dengan basis CMC NA dengan konsentrasi getah pohon jaran 5%, 10%, dan 15%. Hewan coba dalam penelitian ini adalah tikus putih (Rattus norvegicus) jantan berumur 8-12 minggu dengan berat 200-250 gram yang dibagi menjadi 5 kelompok yang masingmasing terdiri dari 4 ekor tikus. Kelompok 1 adalah kelompok kontrol negatif yang dijahit lukanya dengan benang silk steril dan diterapi gentamisin sulfat 0.1%.Kelompok 2 adalah kelompok kontrol positif yang dijahit lukanya dengan benang silk yang dikontaminasi S. aureus 105 CFU/ml. Kelompok 3, 4, dan 5 adalah kelompok yang dijahit lukanya dengan benang silk yang dikontaminasi S. aureusdan diterapi gel getah pohon jaran dengan konsentrasi masing-masing 5%, 10%, dan 15%. Ekspresi TGF-β pada preparat IHK diamati menggunakanimmunoratio. Analisis data dilakukan dengan uji one way ANOVA dan uji tukey. Terapi gel getahpohonjarankonsentrasi 10% menunjukkanpeningkatansignifikanterhadapekspresiTGF-β. Pemberian gel getahpohonjarankonsentrasi 10%menunjukkanhasilterapi yangpaling baikdilihatdariperbaikanjaringanlukaberdasarkanpembentukanjaringangranulasi.p adahistopatologikulit.
English Abstract
Castration is causing open cuts wound in skin. Wound healing process can be contaminated by microorganism. One of the bacteria causing wound infections isStaphylococcus aureus which can make longer time duration of wound healing process. Therefore, effective therapy method is needed to shorten wound healing time with antimicrobial activity. This research is aim to know the effect of Lannea coromandelica gel therapy on TGF-β expression and skin histopathology. The Lannea coromandelica gel was made of CMC NA base with three types concentration 5%, 10%, and 15%. Animal model in this research is male rat (Rattus norvegicus) 200-250 gram aged eight to twelve week-old. The animal model was divided into five group, each group contains of four rats. First group is negative control group, which is suturedwith sterile silk and aseptic methodwith gentamicine sulphate 0.1% therapy. Second group is positive control which is sutured with Staphylococcus aureus 105 CFU/ml contaminated suturewith bacteria.The third, fourth, fifth group were underwent therapy with 5%, 10%, dan 15% Lannea coromandelica gel which is sutured with Staphylococcus aureus 105 CFU/ml contaminated silk. The parameter of this research are TGF-β expressions which are measured by immunohistochemistry and histopatological of wound. The data was analyzed with one way ANOVA testand Tukey’s HSD (Honestly Significance Difference)range test with 95% confidence level (α = 0.05). Therapy of Lannea coromandelica gel concentration of 10% showed a significant increase in TGF-β expressions in the castration model of rat (Rattus norvegicus)skin. Lannea coromandelicagel concentration of 10%shows the results of good therapy seen based on repair of wound tissue through granulation tissue formationfrom histopathology of the skin
