Abstract
Stres merupakan reaksi biologis terhadap rangsangan yang mengganggu homeostatis tubuh baik faktor fisik maupun faktor psikis, bila stimulus yang memicu stres berlangsung lama atau besarnya melebihi kemampuan adaptasi akan menyebabkan gangguan pada sistem saraf dan hormonal. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian diet tinggi lemak terhadap kadar MDA otak besar dan gambaran histopatologi korteks serebri tikus putih (Rattus norvegicus) yang diberi paparan immobilization stress. Tikus di bagi menjadi 4 kelompok dengan kelompok 1 kontrol negatif, kelompok 2 perlakuan stres, kelompok 3 perlakuan diet tinggi lemak, kelompok 4 perlakuan kombinasi (stres dan diet tinggi lemak). Hewan coba tikus di induksi diet tinggi lemak secara sonde lambung sebanyak 5mL/ekor/hari yang dibagi tiga kali pemberian yaitu 2 mL pagi, 1 mL siang dan 2 mL sore. Perlakuan immobilization stress yang di lakukan yaitu 3 jam stres (08.00-11.00), 2 jam istirahat (11.00–13.00), 3 jam stres (13.00- 16.00). Tikus di euthanasi 5 hari setelah perlakuan. Kadar MDA otak besar menggunakan uji ANOVA (Analysis of Variance) dengan taraf kepercayaan sebesar 95% (α = 0,05) dan dilakukan analisis lebih lanjut dengan uji Tukey (α = 0,05). Hasil dari penelitian pemberian diet tinggi lemak terhadap kadar MDA otak besar tikus yang diberi paparan immobilization stress memiliki peningkatan yang signifikan di bandingkan dengan perlakuan immobilization stress dan diet tinggi lemak, sedangkan pemberian diet tinggi lemak terhadap gambaran histopatologi korteks serebri tikus yang diberi paparan immobilization stress terdapat kongesti meninges, kongesti korteks serebri, hemoragi, nekrosis sel dan peradangan. Kesimpulannya pemberian diet tinggi lemak pada tikus yang di papar immobilization stress dapat menyebabkan perubahan pada kadar MDA otak besar serta pemberian diet tinggi lemak pada tikus yang di papar immobilization stress dapat memperparah kerusakan yang terjadi pada histopatologi korteks serebri.
English Abstract
Stress is a biology reaction towards stimulation that interfere body’s homeostatic physically and psychologically. If the stimulation that is triggering the stress take a long time or it’s exceeding the adaptation ability, then it will cause disruption to nervous system and hormones. The objective of this research was to identify the effect of high-fat diet towards the cerebrum MDA levels and histopathologic description on cerebral cortex of rat (rattus norvegicus) given immobilization stress exposure. The rats divided into 4 groups; first group was negative control, second group was immobilization stress treatment, third group was high-fat diet treatment, fourth group is combination treatment (immobilization stress and high-fat diet). The rats were treated by high-fat diet through tube feeding on its side over 5 mL/head/day, which are divided into three treatments; 2 mL in the morning, 1 mL in the afternoon and 2 mL in the evening. Immobilization stress treatments were conducted by this formula: 3 hours stress (08.00 am - 11.00 am), 2 hours break (11.00 am – 01.00 pm), 3 hours stress (01.00 pm - 04.00 pm). Rats were euthanized after 5 days of treatment. The level of cerebrum MDA used ANOVA (Analysis of Variance) test with probably true value 95% (α = 0,05) and further analysis used Tukey test (α = 0,05). The result of induction high-fat diet towards cerebrum MDA levels of the rat given immobilization stress exposure research were has significantly elevation compared to immobilization stress treatment and high-fat diet respectively, while the histopathologic description on rat cerebral cortex were found meninges congestion, cerebral cortex congestion, hemorrhage, cell necrosis and inflammation. In conclusion, the induction of high-fat diet to the rat that is exposed by immobilization stress causes elevation in cerebral MDA levels and make the damage cortex cerebri more severe based on histopathologic description
