Abstract
Diabetes Mellitus adalah penyakit yang ditandai dengan terjadinya hiperglikemia dan gangguan metabolisme karbohidrat, lemak, dan protein yang dihubungkan dengan kekurangan secara absolut atau relatif dari kerja dan atau sekresi insulin. Infeksi bakteri Staphylococcus aureus pada penderita diabetes mellitus dapat memperparah kerusakan jaringan dan terjadi bakteremia. Infeksi S. aureus dapat mengakibatkan bakteremia serta meningkatkan terjadinya inflamasi dan memperparah kondisi karena telah mengalami gangguan metabolisme. Penelitian ini bertujuan untuk mengetahui keadaan tikus putih yang diinduksi streptozotocin dan S. aureus melalui kadar relatif sel T CD4 dan TGF-β. Penelitian ini bersifat eksperimental laboratorik dengan menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari lima perlakuan, yaitu perlakuan 1 (tikus sehat), Perlakuan 2 (diinfeksi S.aureus 108 CFU/mL), dan tiga perlakuan diinduksi streptozotocin dengan dosis 45 mg/kg BB dan diinfeksi S.aureus bertingkat yaitu 105 ,106,107. Induksi streptozotocin single high dose secara intraperitoneal dengan dosis 45 mg/kg BB pada hari kedelapan, pengukuran kadar glukosa darah dilakukan dengan glukometer digital pada hari kesepuluh yang dilanjutkan dengan induksi Staphylococcus aureus secara intraperitoneal. Pengukuran kadar relatif sel T CD4 dan TGF-β dengan menggunakan flowcytometry. Data dianalisis menggunakan uji One Way Analysis of Varience (ANOVA) (α= 0,05). Hasil Penelitian menunjukkan rata-rata kadar relatif CD4 mengalami peningkatan pada perlakuan 3 dan 4 serta rata-rata kadar relatif TGF-β mengalami penurunan pada perlakuan 3,4, dan 5. Kesimpulannya, bahwa kadar relatif CD4 mengalami peningkatan sedangkan kadar relatif TGF-β mengalami penurunan pada tikus putih yang diinduksi STZ dan Staphylococcus aureus.
English Abstract
Diabetes mellitus is a disease that is characterized by the onset of hyperglycemia and impaired metabolism of carbohydrates, fats, and proteins associated with a deficiency in absolute terms or relative from work and secretion of insulin. Staphylococcus aureus infection in animal with diabetes mellitus may be aggravating tissue damage and bakteremia. S. aureus infections can lead to bakteremia as well as increase the occurrence of inflammation and worsen the condition because it has suffered a disturbance of the metabolism. This research aimed to know the state of white rats induced by streptozotocin and S.aureus through the relative levels of CD4 T cells and TGF-β. This research were experimental Completely Randomized Design (RAL), which consists of the five treatment, that treatment 1 (healthy rats), Treatment 2 infection ( S.aureus 108 CFU/mL), and three treatments of streptozotocin-induced with a dose of 45 mg/kg and infect S. aureus multilevel i.e. 105, 106,107. Streptozotocin induced high single dose intraperitoneal doses in 45 mg/kg on the eighth day, the measurement of blood glucose levels was done with digital glukometer on the tenth day of induction followed by Staphylococcus aureus in intraperitoneal. The measurement of the relative levels of CD4 T cells and TGF-β by using flowcytometry. The data were analyzed using One Way Analysis of trials of Varience (ANOVA) (α = 0.05). The results showed an average CD4 levels relatively experienced an increase in treatment 3 and 4 as well as the average relative levels of TGF-β decreased treatment 3.4 and 5. In conclusion, that the relative levels of CD4 had increased while the relative levels of TGF-β decreased in rats induced by streptozotocin and Staphylococcus aureus
