Abstract

Penyakit gangguan reproduksi perlu diketahui dan dipetakan dengan akurat, sehingga upaya pencegahan, pengendalian, serta penanganannya dapat membuahkan hasil yang optimal. OGP (Oviduct-spesific Glycoprotein) merupakan salah satu dari glikoprotein yang memiliki berat molekul tinggi. Glikoprotein ini dihasilkan oleh sel sekretori yang tidak bersilia pada epithelial saluran oviduk yang bersifat estrogen dependen, sehingga terjadinya kasus penyakit reproduksi hipofungsi ovarium, memungkinkan terjadinya perbedaan pada profil pita protein serta kadar protein. Penelitian ini bertujuan untuk mengetahui adanya perbedaan pada profil pita protein dan kadar protein OGP kambing PE betina yang mengalami abnormalitas di ovarium (hipofungsi ovarium). Sampel organ ovarium diperoleh dari limbah RPH kemudian diidentifikasi secara gross anatomy. Analisa profil pita protein dilakukan secara deskriptif, kemudian untuk kadar protein OGP dilakukan secara kuantitatif melalui uji t independen. Hasil penelitian menunjukkan bahwa berat molekul protein OGP pada kambing Peranakan Etawa (PE) betina adalah 62-66 kDa. Dengan pola profil pita protein OGP pada sampel dengan ovarium normal memiliki garis pita lebih tebal apabila dibandingkan dengan sampel organ reproduksi kambing PE yang mengalami hipofungsi. Nilai rata-rata kadar protein OGP antara ovarium normal dan hipofungsi menunjukkan adanya perbedaan yang signifikan (p<0,05), dengan rata-rata kadar OGP pada kelompok ovarium normal sebesar 408.72±38.91 pg dan rata-rata kadar OGP pada kelompok ovarium hipofungsi sebesar 94.85±18.41 pg. Disimpulkan bahwa kondisi ovarium memengaruhi level sekresi protein OGP dalam organ reproduksi betina.

English Abstract

Reproductive disorders need to be known and mapped accurately, so the prevention, control and handling efforts of those disorders can produce optimal results. OGP (Oviduct-specific glycoprotein) is one of the glycoproteins that have high molecular weight. This glycoprotein is produced by nonciliated secretory cells on the oviduct with oestrogen-dependent characteristic, so the occurrence of reproductive hypofunction ovarian disease allows the differences in both of band profile and protein levels. This study aims to determine the differences in OGP band profile and OGP protein levels of female Etawa Crossbreed goats with ovarian abnormalities (ovarian hypofunction). The samples of ovarian organs were obtained from slaughter house waste then identified by gross anatomy. Analysis of protein band profiles was carried out descriptively, then for OGP protein levels carried out quantitatively through an independent t test. The results showed that the molecular weight of OGP protein in female Etawa Crossbreed goats was 62-66 kDa. The pattern of OGP protein band profiles with normal ovaries sample have a thicker band line compared to organs sample with ovarian hypofunction. The mean value of OGP protein levels between normal ovaries and hypofunction showed a significant difference (p <0.05), with an OGP level average in the normal ovary group was 408.72 ± 38.91 pg and the OGP level average in the hypofunction ovary group was 94.85 ± 18.41 pg.
