Abstract
Luka bakar adalah kerusakan atau kehilangan jaringan yang disebabkan oleh panas, radiasi, kimia dan listrik. Luka bakar derajat III merupakan kejadian yang memiliki tingkat keparahan yang tinggi. Lemak omentum sapi diketahui mengandung asam laurat, asam linolenat, asam oleat dan asam arakidonat, yang berfungsi sebaga antimikroba dan berguna dalam penyembuhan luka. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian lemak cair omentum sapi terhadap ekspresi IL-10 dan gambaran histopatologi kulit pada kesembuhan luka bakar derajat III tikus putih (Rattus norvegicus). Penelitian ini menggunakan rancangan acak lengkap dengan 20 ekor tikus putih (Rattus norvegicus) strain Wistar jantan dengan berat badan 100-150 gram dibagi menjadi 4 kelompok dengan 5 ulangan, yaitu K1 (kontrol negatif), K2 (kontrol positif yang diberi terapi dengan Normal Saline), P1 diberi terapi dengan Bioplacenton dan P2 diberi terapi dengan lemak cair omentum sapi steril sebanyak 1 ml diberikan 2 hari sekali selama 14 hari. Pengamatan ekspresi IL-10 menggunakan metode Imunohistokimia dan histopatologi kulit dengan pewarnaan Haematoxylin-Eosin. Data ekspresi IL-10 dianalisa dengan menggunakan uji statistik one-way ANOVA dilanjutkan dengan Uji Beda Nyata Jujur (BNJ) p<0,05 dan histopatologi kulit dianalisa secara deskriptif. Hasil penelitian menunjukkan bahwa pemberian terapi lemak cair omentum sapi secara signifikan (p<0,05) memiliki peningkatan ekspresi IL-10 terbesar dan dapat memperbaiki jaringan epidermis dan dermis pada kulit. Kesimpulan penelitian ini menunjukkan bahwa pemberian terapi lemak cair omentum sapi dapat digunakan untuk penyembuhan luka bakar derajat III dengan meningkatkan ekspresi IL-10 dan memperbaiki gambaran histopatologi kulit
English Abstract
Burn is the loss or damage of tissue caused by heat, radiation, chemicals, and electricity. The third-degree burns had a serious condition. Omental fat contains lauric acids, linolenic acid, oleic acid, and arachidonic acid, which functions as an antimicrobial and contains compounds that are useful in the process of wound healing. The aim of this research was to determine the therapeutic effect of liquid omental fat of cattle toward the expression of IL-10 and skin histopathology of third-degree burns healing on the rat (Rattus norvegicus). This study used a randomized complete design with 20 male white rat (Rattus norvegicus) strain Wistar, weighing around 100-150 gram divided into 4 groups with 5 replication, K1 (negative control), K2 (positive control treated with NS), P1 (treated with Bioplacenton) and P2 (treated with sterilized liquid omental fat of cattle 1 ml/2 days) for 14 days. The observation of IL-10 expression with Immuhohistochemistry staining and skin histopathology in Hematoxylin-eosin staining slides. Data were analyzed with statistical one-way ANOVA followed with Tukey test p<0,05 and descriptive skin histopathology. The results showed that the liquid omental fat of cattle therapy significantly (p< 0,05) could repair the damaged tissue of epidermis and dermis in the skin and increase the expression of IL-10. The conclusion of this study was the liquid omental fat of cattle therapy could be used for the treatment of third-degree burns by increasing the expression of IL-10 and repairing the histopathological image of the skin.
