Abstract

Glomerulonefritis adalah keadaan patologis ginjal yang ditandai pembengkakan glomeruli dan penurunan filtrasi Blood Urea Nitrogen (BUN) dan kreatinin. Streptokinase digunakan untuk induksi glomerulonefritis akut. Kandungan flavonoid pada daun kumis kucing bertindak sebagai antioksidan yang menetralisir radikal bebas sehingga dapat menurunkan kadar BUN dan kreatinin serum. Penelitian bertujuan mengetahui pengaruh terapi ekstrak daun kumis kucing terhadap BUN dan kreatinin serum pada tikus model glomerulonefritis hasil induksi streptokinase. Penelitian menggunakan metode Rancangan Acak Lengkap (RAL) menggunakan 20 tikus jantan berumur 8-12 minggu yang dibagi menjadi 5 kelompok: kontrol negatif yakni tikus sehat, kontrol positif dengan induksi streptokinase 6000 IU, kelompok terapi masing-masing dengan induksi streptokinase 6000 IU dan pemberian ekstrak daun kumis kucing dengan dosis (P1) 250 mg/kgBB, (P2) 500 mg/kgBB, dan (P3) 1000 mg/kgBB. Induksi glomerulonefritis dengan injeksi streptokinase 6000 IU pada vena coccygea dilakukan pada hari ke-1, 6, dan 11. Pengukuran BUN menggunakan metode urease sedangkan kadar kreatinin serum diukur dengan metode Jaffe menggunakan Biosystem Autoanalizer. Data dianalisa dengan one-way ANOVA (a< 0,05) dan uji Tukey. Hasil penelitian menunjukkan bahwa terapi ekstrak daun kumis kucing (Orthosiphon stamineus Benth) secara statistika (P<0,05) menurunkan kadar BUN dan kreatinin, dosis 1000mg/kgBB adalah dosis optimum menurunkan BUN 30,76% dan kreatinin sebesar 85,48%. Disimpulkan bahwa ekstrak daun kumis kucing dapat digunakan sebagai alternatif terapi glomerulonefritis akut berdasarkan kadar BUN dan kreatinin.

English Abstract

Glomerulonephritis is a renal pathological condition characterized by swelling of a glomeruli and decreased filtration ability of Blood Urea Nitrogen (BUN) and creatinine. Streptokinase used for the induction of acute glomerulonephritis. Content of flavonoids in the leaves of Kumis Kucing (Orthosiphon stamineus Benth.) acts as an antioxidant and which neutralizes free radicals to reduce BUN and creatinine serum levels. The aim of the study was to determine the effect of therapeutic extract of kumis kucing leaves on BUN and serum creatinine in streptokinase induced glomerulonephritis models. The study used the Completely Randomized Design (CRD) method. Control design using 20 male rats aged 8-12 weeks divided into 5 groups: negative controls were healthy mice, positive control with 6000 IU streptokinase induction, each treatment group with 6000 IU streptokinase induction and kumis kucing leaf extract therapy with dose (P1) 250 mg/kg BW, (P2) 500 mg/kg BW, and (P3) 1000 mg/kg BW. The induction of glomerulonephritis by injection of 6000 IU streptokinase on the coccygea vein was doing on day 1st, 6th, and 11th. Measurements of BUN using the urease method while serum creatinine levels measured by the Jaffe method using Autoanalizer Biosystem. Data were analyzed by one-way ANOVA (a=0,05) and Tukey test. The results showed that extract of kumis kucing (Orthosiphon stamineus Benth) significally (P <0.05) decreased BUN and creatinine up to 30.76% and 85.48%, respectively with the optimum dose of 1000mg/kg BW. In conclusions leaf extract of kumis kucing can be used as an alternative therapy for acute glomerulonephritis based on BUN and creatinine levels.
