Abstract
Penelitian ini bertujuan untuk mengetahui variasi genetik beberapa spesies cabai (Capsicum spp.) berdasarkan karakter morfologi, kandungan kapsaisin, karotenoid, dan variasi genetik gen Phytoene sintase (Psy). Sampel yang digunakan adalah C. annuum (CAT1 dan CA), C. frutescens (CF), dan turunan Katokkon (KAT). Pada penelitian ini diamati 22 karakter morfologi. Kandungan kapsaisin buah cabai didapatkan dengan metode ekstraksi etanol absolut, sedangkan kandungan karotenoid buah cabai didapatkan dengan metode ekstraksi aseton. Variasi genetik berdasarkan karakter morfologi dan fisiologi dianalisis menggunakan PAST 3, sedangkan variasi gen Psy dianalisis menggunakan MEGA 6.06. Berdasarkan hasil penelitian diketahui variasi morfologi antara CAT1, CA, CF dan KAT dapat dilihat dari karakter pigmentasi nodus, bentuk daun, ukuran daun, warna bunga, bentuk buah, warna buah, ukuran buah, posisi pedicel buah, dan warna biji. Pada umur buah 35-40 HSP kandungan kapsaisin antara CAT1, CA, CF, dan KAT tidak beda signifikan, sedangkan kandungan karotenoid terlihat berbeda signifikan antara kelompok C. annuum (CAT1 dan CA) terhadap CF dan KAT sesuai dengan karakter morfologi warna buah yang teramati. Hasil analisis variasi genetik berdasarkan karakter morfologi dan fisiologi menunjukkan CAT1-CA memiliki nilai koefisien similaritas 93%, sedangkan CF-KAT memiliki nilai koefisien 96%. Variasi molekuler gen Psy mampu membedakan kelompok C. annuum terhadap CF dan KAT. Pohon filogenetik yang dibentuk menunjukkan kedekatan antara CAT1 dan CA termasuk dalam C. annuum, dan kedekatan antara CF dan KAT.
English Abstract
This study aims to determine the genetic variation of several types of chili (Capsicum spp.) based on morphological characters, capsaicin and carotenoid content, and the Pythoene synthase (Psy) gene sequence. The species studied were C. annuum (CAT1 and CA), C. frutescens (CF), and Katokkon (KAT). In this study 22 morphological characters were observed. The capsaicin content of chilli fruit was measured using absolute ethanol extraction method, while the carotenoid content of chilli fruit was measured using acetone extraction method. Genetic variation based on morphological and physiological characters were analyzed using PAST 3, while Psy gene variation were analyzed using MEGA 6.06. It was found that the morphological variations between CAT1, CA, CF and KAT can be seen from the character of nodal pigmentation, leaf shape, leaf size, flower color, fruit shape, fruit color, fruit size, fruit pedicel position, and seed color. At the age of 35-40 fruit HSP the capsaicin content between CAT1, CA, CF, and KAT was not significantly different, whereas the carotenoid content was seen to be significantly different between the C. annuum group (CAT1 and CA) to CF and KAT according to the observed morphological characteristics of the fruit color . The results of the analysis of genetic variation based on morphological and physiological characters show that CAT1-CA has simillarity coefficient 93%, while CF-KAT has simillarity coefficient 96%. The molecular variation of the Psy gene was able to distinguish the C. annuum group from CF and KAT. The phylogenetic tree showed the genetic relationship between CAT1 and CA were include in C. annuum, and genetic relationship between CF and KAT.
