Abstract

Senyawa Klorin (Ca(ClO)2) banyak digunakan sebagai bahan desinfektan dalam air untuk berbagai keperluan termasuk di bidang veteriner melalui klorinasi air. Desinfektan ini bekerja dengan baik untuk membunuh bakteri, fungi dan virus. Namun desinfektan ini juga dapat menimbulkan efek negatif terhadap kesehatan jika terkonsumsi dalam jumlah yang berlebihan. Kemampuan oksidasi senyawa klorin sangat kuat, dalam air klorin dapat diubah menjadi asam hipoklorit (HClO) yang dapat menembus sel dan bereaksi dengan protein sitoplasmik yang dapat merusak struktur sel. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian senyawa klorin berlebih terhadap profil protein dan histopatologi jejenum pada tikus putih (Rattus norvegicus). Pada penelitian ini digunakan 20 ekor hewan coba tikus putih (Rattus norvegicus). Hewan coba dibagi menjadi 5 kelompok, yaitu kelompok kontrol negatif (diberi air minum biasa), dan kelompok perlakuan (tikus diberi larutan klorin dengan konsentrasi klorin dalam air (P1) 50 ppm ; (P2) 100 ppm ; (P3) 150 ppm ; (P4) 200 ppm). Pemberian larutan klorin dilakukan selama 7 hari sebanyak 1 ml setiap harinya. Hasil profil protein dan hasil pengamatan histopatologi jejenum dianalisa secara deskriptif. Profil protein pada jejenum mengalami perubahan setelah diberi paparan senyawa klorin. Ditemukan protein 43 kDa, 75 kDa, dan 60 kDa yaitu protein yang banyak diekspresikan pada jaringan yang mengalami inflamasi. Gambaran histopatologi jejenum menunjukkan adanya perubahan pada bagian vili usus ditandai dengan adanya erosi pada lapisan epitel dan ruptur pada vili serta diikuti adanya infiltrasi sel radang pada lapisan mukosa. Penelitian ini dapat disimpulkan bahwa pemberian senyawa klorin berlebih dapat mempengaruhi perubahan profil protein dan histopatologi jejenum.

English Abstract

Chlorine Compound is widely used as a disinfectant in water for various purpose including in the veterinary field such as chlorination of water. This disinfectant works well to kill bacteria, fungi and viruses. But this disinfectant can also have negative effects on health if consumed in excessive amounts. The oxidation ability of chlorine compound is very strong, in water chlorine can be converted into hypochlorous acid which can penetrate cells and react with cytoplasmic proteins leading to damage cell structure. This study aimed to determine the effect of excess chlorine compound on the protein profiles and histopathology of jejenum in white rats (Rattus norvegicus). In this study 20 animals were tested white rats (Rattus norvegicus). The experimental animals were divided into 5 groups, namely the negative control group (received regular drinking water), and the treatment group (rats were receive chlorine solution with chlorine concentration in water (P1) 50 ppm; (P2) 100 ppm; (P3) 150 ppm; (P4) 200 ppm).The rat received 1 ml of chlorine solution per day for 7 days. The results of protein profiles and histopathology of jejenum were analyzed descriptively. Protein profiles in jejenum changed after exposure to chlorine compounds. Proteins 43 kDa, 75 kDa, and 60 kDa were found, those were widely expressed proteins in inflamation. The histopathological picture of the lesion shows changes in the intestinal villi marked by erosion of the epithelial lining and rupture in the villi and followed by inflammation of the cell infiltration in the lining mucosa. The conclusion is that the administration of excess chlorine compounds can affect changes in protein profile and histopathology of jejenum
