Abstract

Manajemen pemeliharaan dan nutrisi di peternakan rakyat Indonesia yang masih kurang baik menyebabkan terjadinya gangguan reproduksi pada ternak. Salah satu gangguan reproduksi yang mudah ditemui adalah hipofungsi ovarium. Hipofungsi ovarium merupakan abnormalitas pada ovarium yang tidak dapat menjalankan fungsinya dengan normal. Hipofungsi ovarium ditandai dengan folikulogenesis yang terhambat dan kegagalan ovulasi. Tujuan penelitian ini adalah untuk melakukan pengembangan hewan model hipofungsi ovarium dengan induksi salah satu GnRH antagonis yaitu Cetrorelix acetate dan mengamati pengaruhnya terhadap ekspresi reseptor estrogen (ERs) serta perubahan histopatologi pada ovarium. Penelitian ini menggunakan tiga kelompok tikus putih galur Wistar betina, usia 8-9 minggu, dan berat badan 120-160 gram dan dibagi masing-masing 6 ekor. Perlakuan yang diberikan yaitu kelompok kontrol negatif (tanpa paparan Cetrorelix acetate), kelompok perlakuan 1 (paparan Cetrorelix acetate 0,009 mg/ekor), dan kelompok perlakuan 2 (paparan Cetrorelix acetate 0,0135 mg/ekor). Pengamatan ekspresi reseptor estrogen (ERs) pada ovarium dilakukan dengan metode imunohistokimia, dianalisis menggunakan uji ANOVA dan uji lanjutan BNJ (taraf kepercayaan 95%). Pengamatan perubahan histopatologi ovarium dilakukan dengan pewarnaan Hematoksilin-Eosin (HE), kemudian dianalisa secara kualitatif. Hasil dari penelitian ini menunjukkan bahwa kelompok perlakuan P1 dan P2 dengan pemberian GnRH antagonis berbeda signifikan dibandingkan dengan kelompok kontrol negatif. Kelompok perlakuan P2 memiliki penurunan ekspresi reseptor estrogen tertinggi dengan RE-α sebesar 80,7 % dan RE-β sebesar 92,2% dan kelompok perlakuan P1 dan P2 mampu menghambat perkembangan folikel antral yaitu folikel tersier dan folikel de Graaf. Kesimpulan penelitian ini bahwa GnRH antagonis dapat menurunkan ekspresi reseptor estrogen dan menghambat folikulogenesis dalam histopatologi ovarium

English Abstract

Management of livestock and nutrition that still bad led the reproductive disorders in livestock. Reproductive disorder that easily found was ovarian hypofunction. Ovarian hypofunction is an abnormality in the ovary that can’t work properly. Ovarian hypofunction is characterized by inhibition of folliculogenesis and ovulation failure. The aim of this study were to develop an animal model of ovarian hypofunction by induction of GnRH antagonist which is Cetrorelix acetate and to observe its effect on expression of estrogen receptor (ERS) and ovarian histopathology changes. This study used three groups of female white rats, Wistar strain, ages 8-9 weeks, and weigh 120-160 grams and divided into 6 individuals. The treatments given were negative control groups (without exposure of Cetrorelix acetate), group P1 (exposure of Cetrorelix acetate 0.009 mg/head), and group P2 (exposure of Cetrorelix acetate 0.0135 mg/head). Observation of estrogen (ER) receptors in the ovary was carried out by immunohistochemical methods, analyzed by ANOVA and BNJ follow-up tests (95% confidence level). Observation of ovarian histopathology was carried out by Hematoxylin-Eosin (HE) staining, then analyzed qualitatively. The results of this study indicated that the group P1 and P2 with exposure of GnRH antagonists were significantly different compared to the negative control group. Group P2 has the highest estrogen reduction receptor with RE-α was 80.7% and RE-β was 92.2%. Group P1 and P2 inhibited the antral follicles that was tertiary follicle and de Graaf follicle. The conclusion of this study is GnRH antagonists can reduce estrogen receptors and inhibit folliculogenesis in ovarian histopathology.
