Abstract

Kelinci New Zealand (Oryctolagus cuniculus) merupakan salah satu kelinci populer di Indonesia yang sering diperlombakan karena warna rambutnya yang memiliki daya tarik tersendiri. Kelinci New Zealand terbagi menjadi empat berdasarkan warna rambutnya yaitu Kelinci New Zealand Black, Red, Blue, dan White (Albino). Perbedaan warna rambut yang dimilikinya dipengaruhi oleh ekspresi gen Melanocortin 1 receptor (MC1R). MC1R merupakan gen pengkode produksi melanin pada kulit dan rambut. Penelitian ini bertujuan mengetahui adanya perbedaan sekuen gen MC1R pada Kelinci New Zealand dengan metode PCR. Sampel yang digunakan berupa DNA yang diisolasi dari masing-masing satu sampel darah Kelinci New Zealand Black, Red, dan White (Albino). Amplifikasi DNA dilakukan dengan metode PCR menggunakan sepasang primer forward (MC1R_F) 5’- GTG GAC CGC TAC ATC TCC AT -3’ dan reverse (MC1R_R) 5’- TCT CAC CAG GAG CAC AAC AG -3’ yang didesain melalui Primer3Plus menggunakan database NCBI FN658677.1. Analisa sekuen gen dilakukan sekuensing dengan metode Sanger. Hasil sekuen dianalisa menggunakan software Bioedit dan BLAST NCBI. Hasil penelitian menunjukkan sampel Kelinci New Zealand Black terdapat mutasi 2 transisi dan 9 mutasi transversi yang mempengaruhi susunan asam amino, sehingga disebut mutasi miss sense. Pada Kelinci New Zealand Red dan White (Albino) terdapat 1 mutasi transisi pada sekuen ke-369 yang tidak merubah susunan asam amino sehingga disebut silence mutation. Kesimpulan dari penelitian ini adalah terdapat perbedaan urutan basa nukleotida yang mempengaruhi susunan asam amino gen MC1R pada Kelinci New Zealand Black. Sedangkan pada Kelinci New Zealand Red dan White (Albino) perbedaan urutan basa nukleotida tidak mempengaruhi susunan asam amino gen MC1R.

English Abstract

New Zealand rabbit (Oryctolagus cuniculus) is one of the most popular rabbits in Indonesia which is often contested because of the hair color that has its charm. New Zealand rabbits are divided into four by the color of their hair, namely New Zealand Black, Red, Blue and White (Albino) Rabbits. The difference in hair color it possesses is influenced by the expression of the Melanocortin 1 receptor (MC1R) gene. MC1R is a gene that codes for melanin production in the skin and hair. This study aims to determine the differences in the MC1R gene sequence in New Zealand rabbits using the PCR method. The samples used were DNA isolated from each of the New Zealand Black, Red, and White (Albino) Rabbit blood samples. DNA amplification was carried out by PCR method using a pair of forward primers (MC1R_F) 5'- GTG GAC CGC TAC ATC TCC AT-3 'and reverse (MC1R_R) 5'- TCT CAC CAG GAG CAC AAC AG-3' designed through Primer3Plus using Primer3Plus database NCBI FN658677.1. Gene sequence analysis is done by Sanger method. Sequence results were analyzed using Bioedit and BLAST NCBI software. The results showed that the New Zealand Black Rabbit sample contained 2 transitions and 9 transversion mutations that affect the formation of amino acids, so called miss sense mutations. In New Zealand Red and White (Albino) rabbits, there is one transition mutation in the 369th sequence that does not change the composition of amino acids so it is called silence mutation. This study concludes that there are differences in the sequence of nucleotide bases that affect the amino acid composition of the MC1R gene in New Zealand Black Rabbit. Whereas in New Zealand Red and White (Albino) Rabbits the difference in nucleotide base sequence does not affect the amino acid composition of the MC1R gene.
