Abstract
Plumbum merupakan bahan kimia yang berada di lingkungan sekitar dan termasuk salah satu kelompok logam berat yang dapat berbahaya bagi tubuh. Masuknya plumbum ke dalam tubuh dapat melalui saluran pernafasan, pencernaan, dan absorbsi melalui kulit. Akumulasi induksi plumbum terus menerus dapat meningkatkan kadar Reactive Oxygen Species (ROS) dalam tubuh yang akan berdampak pada kerusakan organ terutama jejunum. Kerusakan jejunum dapat menyebabkan peningkatan aktivitas protease jejunum. Ekstrak kulit apel memiliki kandungan flavonoid yang berfungsi sebagai antioksidan dan mampu menetralisir radikal bebas. Penelitian ini bertujuan untuk mengetahui pengaruh terapi preventif Ekstrak Kulit Apel terhadap aktivitas protease dan gambaran histopatologi jejunum tikus putih (Rattus norvegicus) yang diinduksi plumbum. Penelitian ini menggunakan 20 ekor tikus yang dibagi menjadi 5 kelompok perlakuan, yaitu kontrol negatif tanpa diberikan perlakuan, kontrol positif dengan pemberian Pb Asetat 10mg /ekor/hari selama 14 hari, dan kelompok terapi preventif dengan pemberian ekstrak kulit apel 28 mg/200 g BB, 56 mg/200 g BB, 112 mg/200 g BB selama 21 hari dan Pb Asetat dosis 10mg/ekor/hari selama 14 hari pada hari ke-15 sampai hari ke-28. Pengamatan histologi jejunum menggunakan pewarnaan HE yang diamati menggunakan mikroskop cahaya dan pengukuran kadar aktivitas protease menggunakan metode spektofotometri. Aktivitas protease dianalisa menggunakan analisa ragam ANOVA menggunakan software SPSS (Statistical Package for Social Sciences) dan dilanjutkan uji BNJ (α = 5%). Hasil penelitian menujukkan pemberian terapi preventif ekstrak kulit apel varietas Rome Beauty dapat menurunkan aktivitas protease dan memperbaiki kerusakan organ jejunum dengan dosis terbaik yaitu 112 mg/200 g BB. Kesimpulan dari penelitian ini adalah ekstrak kulit apel Rome Beauty dapat digunakan sebagai terapi preventif untuk menurunkan radikal bebas akibat induksi plumbum asetat.
English Abstract
The entry of plumbum into the body can be through inhalation, digestion, and absorption through the skin. Continuous accumulation of plumbum induced can increase the number of Reactive Oxygen Species (ROS) which will have an impact to organ damage, especially jejunum. Jejunal damage can cause the increasing jejunal protease activity. Apple peels extract contains flavonoids which function as antioxidants to neutralize free radicals. The aims of this research were to identify the effect of apple peels extract as preventive therapy on protease activity and jejunal histopathology plumbum-induced of rat (Rattus norvegicus). This research used 20 rats, those were divided into 5 treatment groups, which were negative control group without treatment, positive control group which induced Pb acetate 10 mg/rat/day for 14 days, and preventive therapy group which administrated the apple peels extract with 28 mg / 200 g BW, 56 mg / 200g BW, 112mg / 200g BW doses for 21 days and also Pb acetate with 10mg/rat/day for 14 days on day 15th until 28th. Jejunal histological features was stained using HE stain which then observed using a microscope and protease activity levels measured using by the spectrophotometric method. Protease activity was analyzed using a variety of ANOVA analyzes using SPSS software (Statistical Package for Social Sciences) and continued by Tukey test (α = 5%). The results showed that the administration of Rome Beauty varieties apple peels extract as preventive therapy with the best dosage of 112 mg / 200 g BW can reduce protease activity and improve jejunal histopathology. The conclusion of this study is that apple peels extract of Rome Beauty can be used as a preventive therapy to reduce free radicals due to plumbum acetate induced.
