Abstract

Hiperkolesterolemia adalah suatu kondisi saat kolesterol di dalam darah meningkat melebihi batas normal. Ekstrak kulit pisang kepok (Musa paradisiaca) mengandung pektin, tannin, saponin, dan flavonoid yang memiliki efek preventif terhadap hiperkolesterolemia. Penelitian ini bersifat True Experimental menggunakan Rancangan Acak Lengkap (RAL), dengan 20 ekor tikus putih (Rattus norvegicus) jantan strain Wistar, yang terbagi dalam lima kelompok yaitu kelompok kontrol negatif (K-), kontrol positif (K+), perlakuan 1 (P1), perlakuan 2 (P2), dan perlakuan 3 (P3). Ekstrak kulit pisang kepok (Musa paradisiaca) diberikan secara preventif dengan dosis 22,05mg/150gBB (P1), 44,1mg/150gBB (P2), dan 88,2mg/150gBB (P3) selama 1 minggu dan dilanjutkan selama 3 minggu secara bersamaan dengan diet hiperkolesterolemia dengan jeda waktu satu jam. Pemberian diet hiperkolesterolemia pada kelompok K+, P1, P2, dan P3 dengan komposisi asam kholat 0,1%, minyak babi 10%, dan kuning telur puyuh 5% yang diberikan selama 3 minggu dengan dosis 3,02g/ekor. Analisis data kuantitatif berupa kadar kolesterol total menggunakan uji ANOVA (α=0,05) dan dilanjutkan dengan uji Tukey (p<0,05). Data kualitatif berupa gambaran histopatologi duodenum yang dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa semua dosis pemberian ekstrak kulit pisang kepok (Musa paradisiaca) mampu mencegah peningkatan kadar kolesterol total dan mencegah erosi epitel, hipertrofi sel goblet, dan infiltrasi sel radang pada gambaran histopatologi tunika mukosa duodenum. Kesimpulan bahwa dosis 88,2mg/150gBB merupakan dosis efektif untuk mencegah peningkatan kadar kolesterol total dan mencegah kerusakan histopatologi duodenum pada tikus model hiperkolesterolemia.

English Abstract

Hypercholesterolemia is a condition when cholesterol in the blood increase over the normal limit. Musa paradisiaca peel extract contains of pectin, tannin, saponin, and flavonoid which have a preventive effect on hypercholesterolemia. This study is True Experimental using Completely Randomized Design (CRD), with 20 rats (Rattus norvegicus) male Wistar strains divided into five groups : negative control (K-), positive control (K+), treatment 1 (P1), treatment 2 (P2), and treatment 3 (P3). Musa paradisiaca peel extract was given preventive with dose 22.05mg/150gBW (P1), 44.1mg/150gBW (P2), and 88.2mg/150gBW (P3) for 1 weeks and continued for 3 weeks together with a diet of hypercholesterolemia with interval of one hour. The diet of hypercholesterolemia was given for K+, P1, P2, and P3 groups with composition 0.1% cholic acid, 10% pork oil, and 5% quail egg yolk, for 3 weeks with dose 3.02g /rat. Quantitative data analysis of total cholesterol levels were analyzed using the ANOVA test (α=0.05) and continued with the Tukey test (p <0.05). Qualitative data of duodenum histopathology were analyzed descriptively. The results showed that all doses of Musa paradisiaca peel extract were able to prevent an increase total cholesterol levels and prevent epithelial erosion, goblet cell hypertrophy, and infiltration inflamatory cells in tunica mucosa of duodenum histopathology. The conclusion that the dose of 88,2mg/150gBW is an effective dose to prevent an increase total cholesterol levels and prevent duodenum histopathology damage in rats model of hypercholesterolemia.
