Abstract

Owa Jawa (Hylobates moloch) merupakan primata endemik yang hanya ditemukan di Pulau Jawa, Indonesia. Status owa Jawa di IUCN-World Concervation yaitu terancam punah (Endangered) dan Apendik II CITES. Penelitian mengenai analisa kekerabatan antar individu owa Jawa menggunakan DNA mitokondria (mtDNA) pada gen Cytochrome C Oxidase subunit 1 (COI) perlu dilakukan demi mendukung upaya pelestariannya. Gen COI berukuran 228 bp diperoleh dari sampel berupa rambut dari 3 individu owa Jawa jantan setelah melalui tahap isolasi DNA menggunakan Geneaid DNA Mini Kit dan amplifikasi dengan metode Polymerase Chain Reaction (PCR) menggunakan sepasang primer forward (OJW_F) 5'-ATG TCG TAG CCC ACT TCC AC-3' dan reverse (OJW_R) 5'-TGT ATG CAT CGG GGT AGT CA-3'. Sekuens hasil PCR dilakukan dengan metode Sanger. Hasil sekuen gen dianalisa secara deskriptif kualitatif untuk memperlihatkan kekerabatan antar sampel individu owa Jawa (Hylobates moloch) dengan data dari NCBI dengan kode akses HQ622784.1 (intraspesies), lalu dibandingkan dengan owa Hoolock (Hoolock hoolock) kode akses KY250073.1 dan Siamang (Symphalangus syndactylus) kode akses HQ622798.1 (interspesies) menggunakan program BioEdit®, Basic Local Aligment Search Tool (BLAST) dari NCBI, dan program Molecular Evolutionary Genetics Analysis (MEGA). Hasil penelitian menunjukkan bahwa kekerabatan owa Jawa yang berada di Javan Primate Rehabilitation Center (JPRC) Jawa Barat berhasil dideskripsikan berdasarkan Gen COI (Cytochrome C oxidase subunit 1) secara intraspesies maupun interspesies. Jarak genetik intraspesies sampel owa Jawa yang dibandingkan dengan sekuen gen COI owa Jawa database NCBI menunjukkan kekerabatan yang sangat dekat karena bernilai sama atau kurang dari 1%. Jarak genetik interspesies yang dibandingkan dengan Siamang (Symphalangus syndactylus) genus Symphalangus bernilai 8,6%, sedangkan jika dibandingkan dengan owa Hoolock (Hoolock hoolock) genus Hoolock bernilai 5,8% sampai 7,9%. Hal ini menunjukkan bahwa owa Jawa secara molekuler lebih dekat dengan genus Hoolock daripada genus Symphalangus

English Abstract

Javan gibbon (Hylobates moloch) is an endemic primate found only on Java, Indonesia. Status of Javan gibbons at the IUCN-World Conservation, is endangered and Appendix II at CITES. Research on kinship analysis between Javan gibbon individuals using mitochondrial DNA (mtDNA) on the Cytochrome C Oxidase subunit 1 (COI) gene needs to be done to support its conservation efforts. The COI gene measuring 228 bp was obtained from hair samples of 3 male Javan gibbons after going through the DNA isolation stage using Geneaid DNA Mini Kit and amplification using the Polymerase Chain Reaction (PCR) method using a pair of forward primers (OJW_F) 5’-ATG TCG TAG CCC ACT TCC AC-3 ‘and reverse (OJW_R) 5’-TGT ATG CAT CGG GGT AGT CA-3’. The sequence of results of PCR was carried out by the Sanger method. The results of gene sequences were analyzed qualitatively to show the relationship between individual samples of Javan gibbons (Hylobates moloch) and data from NCBI with access codes HQ622784.1 (intraspecies), then compared to Hoolock gibbons (Hoolock hoolock) access codes KY250073.1 and Siamang (Symphalangus syndactylus) access code HQ622798.1 (interspesies) using the BioEdit® program, Basic Local Aligment Search Tool (BLAST) from NCBI, and programs Molecular Evolutionary Genetics Analysis (MEGA). The results showed that the kinship of Javanese gibbons in the Javan Primate Rehabilitation Center (JPRC) of West Java was successfully described based on the COI gene (Cytochrome C oxidase subunit 1) both intraspecifically and interspecies. The genetic distance between the species of Javan gibbon compared to the COI gene sequences of Javanese gibbons in the NCBI database shows a very close relationship because it is worth the same or less than 1%. The interspecies genetic distance compared to the Siamang (Symphalangus syndactylus) genus Symphalangus is worth 8.6%, whereas when compared with the Hoolock (Hoolock hoolock) genus the Hoolock genus is worth 5.8% to 7.9%. This shows that Javan gibbons are molecularly closer to the genus Hoolock than the genus Symphalangus.
