Abstract

Salah satu penyakit pencernaan adalah Inflammatory Bowel Disease (IBD). Inflammatory Bowel Disease dapat disebabkan oleh berbagai macam faktor seperti efek negatif Obat Anti Inflamasi Non Steroidal (OAINS) seperti indometasin. Salah satu tanaman yang dimanfaatkan sebagai obat tradisional untuk terapi penyakit adalah tanaman sukun yang merupakan tanaman asli Indonesia di mana daunnya telah digunakan secara tradisional dalam pengobatan berbagai macam penyakit contohnya seperti penyakit pencernaan. Tujuan dari penelitian ini adalah untuk mengetahui efek terapi ekstrak daun sukun (Artocarpus altilis) terhadap inflammatory bowel disease (IBD) pada tikus putih (Rattus novergicus) berdasarkan kadar Malondialdehyde (MDA) dan Superoxide Dismutase (SOD) pada duodenum. Penelitian ini menggunakan rancangan acak lengkap dengan 20 ekor tikus putih (Rattus norvegicus) strain Wistar jantan umur 12 minggu dengan berat badan 150-250 gram dibagi menjadi 5 kelompok, yaitu K1 (kontrol negatif), K2 (kontrol positif yang diinduksi 15mg/kg BB indometasin), K3, K4, dan K5 merupakan kelompok terapi ektrak daun sukun dosis 100 mg/kg BB, 200 mg/kg BB, 400 mg/kg BB tikus putih yang sebelumnya telah diinduksi indometasin 15mg/kgBB. Induksi indometasin dilakukan satu kali pada hari ke 8. Terapi ekstrak daun sukun dengan cara sonde lambung dimulai dari hari ke 9 sekali sehari selama 14 hari. Pengukuran kadar MDA dilakukan dengan metode TBA dan pengukuran kadar SOD dilakukan dengan metode NBT, kemudian dianalisis dengan metode statistik one way ANOVA. Hasil penelitian ini adalah pemberian esktrak daun sukun (Artocarpus altilis) dengan dosis 400 mg/kgBB lebih efektif menurunkan kadar MDA, sedangkan dosis 200mg/kgBB dan 400mg/kgBB efektif untuk meningkatkan kadar SOD pada tikus model Inflammatory Bowel Disease.

English Abstract

Inflammatory Bowel Disease (IBD) is One of the digestive diseases. Inflammatory Bowel Disease can be caused by various factors such as the negative effects of Non-Steroidal Anti-Inflammatory Drugs (NSAIDs) such as indomethacin. The plant used as traditional medicine for the treatment of diseases is breadfruit plants which are native to Indonesia where the leaves have been used traditionally in the treatment of various diseases, such as digestive diseases. The purpose of this study was to determine the therapeutic effects of breadfruit leaf extract (Artocarpus altilis) on inflammatory bowel disease (IBD) in white rats (Rattus novergicus) based on levels of Malondialdehyde (MDA) and Superoxide Dismutase (SOD) in the duodenum. This study used a completely randomized design with 20 male white rats (Rattus norvegicus) of Wistar strain aged 12 weeks with a weight of 150-250 grams divided into 5 groups, namely K1 (negative control), K2 (positive control induced 15 mg / kg BW indomethacin), K3, K4, and K5 were groups of breadfruit leaf extract therapy dose of 100 mg / kg, 200 mg / kg body weight, 400 mg / kg body weight white rats after induced 15mg/kgBW indomethacin. Indomethacin induction was done once on the 8th day. Breadfruit leaf extract therapy by method of gastric sonde starts from day 9 once a day for 14 days. MDA level measurement was carried out by the TBA method and SOD level measurement was carried out using the NBT method, then analyzed by oneway ANOVA statistical methods. The results of this study are the administration of breadfruit leaf extract (Artocarpus altilis) at a dose of 400 mg / kgBB more effective than MDA levels, while the dosages of 200mg / kgBW and 400mg / kgBB are effective for increasing SOD levels in white rats models of Inflammatory Bowel Disease.
