Abstract

Sapi Peranakan Friesian Holstein (Bostaurus) merupakanmamaliaendemikPulauJawa yang memilikiperbedaankualitas semen padasetiapindividu. Tyrosin kinase (TEK) merupakansalahsatu gen yang menentukankualitas semen. Protein Tyrosin kinase ialahsalahsatumolekul protein yang terdapatpadamembran plasma spermatozoa danberfungsipengenalandengan ZP3 sertaberperandalam signal transduksi yang akanmenghasilkanautofosforilasi. Penelitianinibertujuanuntukmengidentifikasipolimorfismepada Gen TEK gunamendukung proses seleksipejantanberdasarkansekuen gen TEK metode PCR. Sampel yang digunakanadalah DNA yang diisolasidaridarah. Amplifikasi DNA dilakukandenganmetode PCR menggunakan primer Forward (TEK_F) 5’- TAGATTGTCGCTTGCCTGGG-3’ danReverse (TEK_R) 5’- CCTGTGCCGACAGGTTTACT-3’ dandilanjutkandengansekuensingterhadapproduk PCR. Analisahasilsekuen DNA dilakukandenganmenggunakan software BioEditdan NCBI BLAST. Hasilpenelitianmenunjukkanbahwadari ke-7 sampel menghasilkan 262 bp dan ditemukan polimorfisme/perubahan sekuengen TEK pada 23databasegene bank. Padaanalisishubunganantaramotilitas individu spermatozoadenganmutasi, diperolehrhitung>rtable (0,806 > 0,754) atau nilai signifikansi< taraf nyata 5% (0,029< 0,050). Padaanalisishubunganantarakonsentrasisemen denganmutasi, diperolehrhitung>rtable (0,897 > 0,754) ataunilaisignifikansi<tarafnyata 5% (0,006 < 0,050).

English Abstract

The Holstein Friesian Crossbreed bull (Bostaurus) is an endemic mammal of Java Island that has an amount of semen quality in each individual. Tyrosine kinase (TEK) is one of the genes that determine semen quality. Protein Tyrosine kinase is one of the proteins present in the plasma membrane and works with ZP3 and signals in signal transduction which will produce autophosphorylation. This study aims to identify polymorphisms in TEK genes to support the male selection process based on TEK gene sequences by the PCR method. The DNA sampels were isolated from the blood. The process of DNA amplification was carried out using the PCR method using the Forward (TEK_F) primer 5'-TAGATTGTCGCTTGCCTGGG-3 'and Reverse (TEK_R) 5'- CCTGTGCCGACAGGTTTACT-3' and continued by sequencing of PCR products. The DNA sequence was carried out using BioEdit software and NCBI BLAST. The results showed that from the 7 samples produced 262 bp and found a polymorphism/changes in the sequence of the TEK gene on 23 database gene bank. On the analysis of the relationship between individual motility of spermatozoa and mutations, r count> r table (0.806> 0.754) or significance value <5% (0.029 <0.050) was obtained. In analyzing the relationship between cement concentration and mutation, r count> r table (0.897> 0.754) or significance value <5% (0.006 <0.050) was obtained.
