Abstract

Indonesia masih memiliki daerah endemik penyakit malaria. Plasmodium falciparum merupakan parasit yang menjadi penyebab utama penyakit malaria. Daun Mimosa pudica mengandung flavonoid, terpenoid, dan alkaloid yang berpotensi sebagai antimalaria. Penelitian ini bertujuan untuk mengetahui ada tidaknya perbedaan aktivitas antimalaria ekstrak etanol 96% daun M. pudica dengan klorokuin. Plasmodium dikultur menggunakan metode Trager dan Jensen yang telah dimodifikasi. Persen parasitemia awal yang digunakan untuk pengujian adalah 1%. Persen parasitemia dihitung secara visual dengan mikroskop cahaya. Persen hambatan dihitung dengan membandingkan persen pertumbuhan dengan kontrol negatif. Aktivitas antimalaria ditentukan dengan nilai IC50. Hasil korelasi Spearman antara konsentrasi ekstrak dengan persen parasitemia dan persen hambatan menunjukkan korelasi yang kuat (p = 0,000) dengan nilai r berturut-turut -0,960 dan 0,960. Hasil korelasi Spearman antara konsentrasi klorokuin dengan persen parasitemia dan persen hambatan menunjukkan korelasi yang kuat (p = 0,000) dengan nilai r berturut-turut -0,985 dan 0,985. Semakin tinggi konsentrasi ekstrak, maka persen parasitemia semakin menurun dan persen hambatan pertumbuhan semakin meningkat. Berdasarkan analisis probit, dapatkan nilai IC50 ekstrak sebesar 0,080 ± 0,045 μg/mL dan IC50 klorokuin 0,062 ± 0,08 μg/mL. Tidak terdapat perbedaan antara aktivitas antimalaria ekstrak etanol M. pudica dengan klorokuin (t test, p = 0,754). Dapat disimpulkan bahwa ekstrak etanol M. pudica memiliki aktivitas antimalaria terhadap Plasmodium falciparum 3D7 .

English Abstract

There are several endemic areas of malarial infection in Indonesia. Plasmoodium falciparum is the main cause of malarial infection. Mimosa pudica leaves contain flavonoids, terpenoids, and alkaloids that have the potential to be developed as antimalarial agents. The aim of this study was to determine antimalarial activity of ethanol extract of M. pudica leaves compared to chloroquine. Plasmodium culture was carried out using Trager and Jensen with modification. P. falciparum with 1% initial parasitemia percentage was used as antimalarial test. Parasitemia percentage was observed under light microscope. Inhibition percentage and IC50 were calculated. Result of Pearson correlation of extract concentrations between percent parasitemia and percent inhibition showed a strong correlation (p = 0.000) with r values -0.960 and 0.960 respectively. Result of Pearson correlation of chloroquine concentration between percent parasitemia and growth inhibition showed a strong correlation (p = 0.000) with r values -0.985 and 0.985 respectively. The values of percent parasitemia decrease as the values of concentration increase while the values of percent inhibition increase as the values of concentration increase. Based on probit analysis, IC50 M. pudica extract was 0.080 ± 0.045 μg/mL and IC50 chloroquine was 0.062 ± 0.08 μg / mL. There was no difference of antimalarial activity between extract and chloroquine (t test, p = 0.754). It can be concluded that M. pudica extract has antimalarial activity against P. falciparum 3D7.
