Abstract
Tanaman melon (Cucumis melo L.) merupakan tanaman semusim yang banyak dibudidayakan di Indonesia dan mempunyai prospek pasar yang menjanjikan. Namun jika dilihat dari produksi buah melon dari tahun ke tahun terus mengalami fluktuasi. Hal ini dikarenakan dalam kegiatan budidaya tanaman melon seringkali mengalami kendala seperti serangan penyakit yang sering kali merugikan petani. Salah satunya adalah penyakit layu fusarium yang disebabkan oleh patogen Fusarium oxysporum f.sp. melonis. Patogen ini menyerang hampir di seluruh fase tanaman melon terutama jaringan bagian vaskuler dan menghambat aliran air pada jaringan xylem sehingga menyebabkan kelayuan pada tanaman inangnya. Sedangkan pengedalian dengan fungisida sisntetik tidak memberikan hasil yang memuaskan. Oleh karena itu, perlu dilakukan pengendalian alternatif yang aman dan ramah lingkungan. Seperti dengan memanfaatkan mikroorganisme yang bersifat antagonis terhadap patogen tersebut, salah satunya adalah dengan memanfaatkan khamir. Penelitian dilaksanakan di Laboratorium Penyakit Tumbuhan, Jurusan Hama dan Penyakit Tumbuhan, Fakultas Pertanian, Universitas Brawijaya pada Februari sampai dengan Agustus 2016. Eksplorasi khamir diambil dari tanah perakaran tanaman melon di UPT Lebo Sidoarjo, serta uji antagonis khamir yang diperoleh terhadap F. oxysporum f.sp. melonis pada media PDA. Khamir yang didapatkan dari tanah perakaran tanaman melon yang berhasil diisolasi berjumlah 6 jenis diantaranya adalah Candida sp., Pichia sp., Zygosaccharomyces sp., Cryptococcus sp., Hanseluna sp., Metschnikowia sp. Persentase penghambatan tertinggi ditunjukkan pada perlakuan Hansenula sp. sebesar 39,64%, sedangkan persentase penghambatan terendah ditunjukkan pada perlakuan Zygosaccharomyces sp. sebesar 11,68%.
English Abstract
Melon (Cucumis melo L.) is one of the popular seasonal fruits in Indonesia has a very promising market. Despite its popularity, statistics shown that the production is fluctuated year by year. One of the reason is fusarium wilt disease which often infect melon plants. One of them is fusarium wilt caused by Fusarium oxysporum f.sp melonis, attacks almost all phases of the plant, especially the vascular tissue and inhibits the flow of water in the xylem tissue, causing the host plant to wilt. Meanwhile, disease control with synthetic fungicide does not provide satisfactory results. Therefore, a safe and ecological friendly alternative control need to be done. One of them is by using antagonist micoorganisms, by using yeast against the pathogen. The research was conducted at the Laboratory of Plant Disease, Department of Plant Pests and Diseases, Faculty of Agriculture, Brawijaya University in February to August 2016. Yeast exploration was taken from the root area of melon plants in UPT Lebo Sidoarjo and from yeast antagonist tests that were obtained against F. oxysporum f.sp. melonis on PDA media. There were 6 types of yeast from the soil in the rizhosfer of melon plants that were successfully isolated, including Candida sp., Pichia sp., Zygosaccharomyces sp., Cryptococcus sp., Hansenula sp., Metschnikowia sp. The highest inhibition percentage is shown in Hansenula sp. with 39.64% amount, while the lowest inhibition percentage shown in Zygosaccharomyces sp. with a number of 11.68%.
